摘 要:为了解乌裕尔河流域富裕段麦穗鱼感染吸虫囊蚴的种类,本研究采集乌裕尔河流域富裕段麦穗鱼,利用人工消化法收集的不同形态的吸虫囊蚴,在显微镜下观察,根据囊蚴的形态特点进行分类。同时分别提取各种囊蚴的总DNA,利用PCR方法扩增其第二内转录间隔区(ITS2)基因序列并进行测序,将获得的基因序列与Gen Bank数据库相应的基因序列进行比对分析,构建系统发生树。结果显示乌裕尔河流域富裕段麦穗鱼主要感染3种吸虫囊蚴,分别为华支睾吸虫、东方次睾吸虫和日本全冠吸虫。本研究进一步证明形态学结合分子生物方法是鉴定淡水鱼中吸虫囊蚴感染种类的有效手段。In order to investigate the kinds of the trematode metacercariae infected Pseudorasbora parva in Fuyu region of Wuyuer river basin, P.parva were collected in this region and the trematode metacercariae were isolated using artificial digestion method. The trematode metacercariae were classified by morphological characteristics utilizing the microscope. The total DNA of each kind of metacercariae was extracted, and the second internal transcriptional space (ITS2) sequences were amplified by PCR and sequenced. Sequences were assembled and aligned with the sequences available in GenBank database, and the phylogenetic relationships were reconstructed based on ITS2 sequences with MP methods. The results showed that three kinds of metacercariae were identified in P.parva, named Clonorchis sinensis, Metorchis orientalis and Holostephanus dubinini, respectively. It is suggested that morphology combined with molecular biological methods is an effective method for identification of metacercariae in freshwater fish.
