摘 要:目的制备盐酸黄酮哌酯缓释片,并对其处方进行优化。方法以羟丙甲基纤维素(HPMC)为基本骨架材料,用湿法制粒压片法制粒压片。应用星点设计-效应面法,以各指标的总评"归一值"为因变量,采用二项式拟合建立因素与因变量的数学关系,根据模型绘制效应面图和等高线图优化处方并进行验证。结果最优处方组成:HPMC-4000为68.44 mg,HPMCAS为4.60 mg,糖粉为19.60 mg。结论盐酸黄酮哌酯缓释片的制备简单易行,星点设计-效应面法建立的数学模型具有准确的预测性。Objective The purpose of this study was to prepare sustained-release tablets of flavoxate hydrochloride and optimize their formulation. Methods Flavoxate hydrochloride sustained-release tablets were prepared by wet granulation technique with the Hydroxypropyl methylcellulose( HPMC) as the basic skeleton material. Applying the central composite design response surface methodology,the overall desirability of each index was chosen as dependent variable. The mathematic relation between dependent variables and independent variables was established by second-order quadratic model. The prescription was optimized and verified by drawing the response surface and contour plots according to the model. Results The best formulation composition was as follow: 68. 44 mg of HPMC-4000,4. 60 mg of HPMCAS,and19. 60 mg of sugar. Conclusion The preparation of flavoxate hydrochloride sustained-release tablets is simple and practicable. The mathematic model established by central composite design-response surface methodology has an accurate predictability.
