摘 要:目的采用HPLC法,建立苦参中氧化苦参碱、苦参碱,藜芦中藜芦胺的含量测定方法,研究苦参单煎液、藜芦单煎液与不同配伍比例合煎液中3种成分的变化规律,从化学物质基础层面,初步阐述苦参藜芦的配伍禁忌机制。方法制备苦参单煎液、藜芦单煎液与不同配伍比例合煎液,采用Wonda Sil C18-WR色谱柱(250 mm×4.6 mm,5μm),以K_2HPO_4·3H_2O的水溶液(浓度20 mmol·L^(-1),H_3PO_4调pH至7.10)为流动相A,乙腈为流动相B,梯度洗脱。检测波长:220 nm,进样量:20μL,柱温:30℃,流速:1.0 m L·min^(-1),依次进样分析。结果氧化苦参碱、苦参碱和藜芦胺质量浓度分别在30.00~960.00 mg·L^(-1)、15.00~480.00 mg·L^(-1)、12.50~400.00 mg·L^(-1)内与峰面积呈良好的线性关系,相关系数分别为0.999 9、0.999 9、0.999 6;苦参藜芦(质量比为1∶1)合煎液中,三者的平均回收率分别为99.4%、98.6%和97.2%;RSD分别为0.9%、1.1%和1.2%(n=6)。结论建立的方法适用于研究苦参单煎液、藜芦单煎液与不同配伍比例合煎液中氧化苦参碱、苦参碱和藜芦胺的变化规律。Objective To establish an HPLC method for determination of oxymatrine,matrine in Sophora flavescent and veratramine in Veratrum nigrum,which was used to study the variation of three constituents in S. flavescens decoction,V. nigrum decoction and combinations of S. flavescens with V. nigrum at different compatibility ratios. To describe the incompatibility mechanism in the combination of S. flavescens and V.nigrum from the basic chemical substances. Methods S. flavescens decoction,V. nigrum decoction and combinations of S. flavescens with V. nigrum at different compatibility ratios were prepared. The separation was developed on a Wonda Sil C18-WR column( 250 mm × 4. 6 mm,5 μm) by gradient elution with acetonitrile-water( containing 20 mmol·L^-1 K2HPO4·3H2O,with H3PO4 adjusted pH to 7. 10) as the mobile phase,flowrate at 1. 0 mL·min^(-1),detection wavelength at 220 nm and column temperature at 30 ℃. In this case,samples were analyzed sequentially. Results The calibration curve was linear within the range of 30. 00-960. 00 mg·L^-1( r = 0. 999 9),15. 00-480. 00 mg·L^-1( r = 0. 999 9),12. 50-400. 00 mg·L^-1( r =0. 999 6) for oxymatrine,marine and veratramine,respectively. In combination of S. flavescens with V.nigrum( m∶ m = 1∶ 1),the average recoveries were 99. 4%( RSD = 0. 9%),98. 6%( RSD = 1. 1%) and97. 2%( RSD = 1. 2%) for oxymatrine,marine and veratramine,respectively. Conclusion The method can be used for the variation of oxymatrine,marine and veratramine in S. flavescens decoction,V. nigrum decoction and combinations of S. flavescens with V. nigrum at different compatibility ratios.
