摘  要:目的探讨采用密度梯度离心法和差速贴壁法分离兔骨髓源性血管内皮祖细胞,并对分离细胞进行培养及鉴定。方法通过细胞形态观察、免疫荧光染色、管腔形成实验以及低密度脂蛋白吞噬和植物凝集素贴附实验进行鉴定。结果分离培养的细胞呈多角形生长,细胞能够形成放射样克隆集落,细胞表达血管假性血友病因子(vW F),在基质胶呈管腔样生长,吞噬低密度脂蛋白并能够黏附植物凝集素。结论采用密度梯度离心法和差速贴壁法分离兔骨髓源性干细胞,在一定条件下能分化成为血管内皮祖细胞。Objective To explore the separation of rabbit bone marrow-derived vascular endothelial progenitor cells(EPCs) with the density gradient centrifugation and differential adhesion methods. Methods The separated cells were cultured and identified. EPCs were identified by general cellular morphologic observation, vW F immunofluorescence, lumen formation experiment, low density lipoprotein phagocytosis, and plant agglutinin attachment test. Results The EPCs grew as polygons, and could form clone colony-like radiation. The vW F was expressed in EPCs. The EPCs grew in the Matrigel as lumen, which can phagocytize low density lipoproteins and adhere to plant agglutinin. Conclusion It was concluded that the isolated rabbit bone marrow-derived monocytes cells which were separated through density gradient centrifugation and differential adhesion methods can successfully differentiate into endothelial progenitor cells under certain conditions.
