摘 要:目的建立用于检测人血浆中伏立康唑浓度的HPLC法。方法以氯唑沙宗为内标,色谱柱为Agilent Zorbax Eclipse Plus C18(100 mm×4. 6 mm,3. 6μm);流动相为乙腈-0. 01 mol·L-1磷酸二氢钾溶液(35∶65);流速0. 8 m L·min-1;柱温35℃;检测波长260 nm。结果伏立康唑在质量浓度0. 2~20 mg·L-1内线性关系良好(r=0. 999 9),定量下限为0. 2mg·L-1,日间、日内精密度良好(RSD均<10%),低、中、高质量浓度下的相对回收率分别为104. 84%、99. 30%、100. 36%,提取回收率> 90%。结论本方法操作简便,结果准确,适用于人血浆中伏立康唑浓度的检测。AIM To establish an HPLC method for the determination of the concentration of voriconazole in human plasma. METHODS Chlorzoxazone was used as the internal standard. The analytical column was packed with Agilent Zorbax Eclipse Plus C ls (100mm×4.6mm,3.6μm) , the mobile phase was acetonitrile -0.01 mol · L-1 monopotassium phosphate (35:65) ,the flow rate was 0.8 mL ·min-1 ,the temperature was 35 ℃ and the de- tection wavelength was 260 nm. RESULTS The liner range of voriconazole in human plasma was 0.2 - 20 mg · L - 1 ( r = 0. 999 9). The limit of quantitation was 0.2 mg · L - 1. The intra-day and inter-day RSD for assaying the plasma sample containing three concentration of voriconazole were both lower than 10%. The relative recovery was 104.84% , 99.30% and 100.36% respectively ,and the extraction recovery was more than 90%. CONCLUSION The method described is simple and accurate for the determination of the concentration of voriconazole in human plasma.
