摘 要:目的应用Taqman探针法检测淡水溺死后大鼠肺组织和血清中CCL2、CCL7以及CCR2 mRNA的表达水平,从而探讨其在法医学实践中,是否能为溺死的诊断提供辅助的科学依据。方法 24只健康雄性SD大鼠随机分为实验组(淡水溺死组)、对照组(空白对照组、淡水应激组及疾病对照组)。对各组大鼠尸表及内部脏器进行肉眼观察并取部分左心室心肌和右下叶肺组织行HE染色;提取大鼠右心室血清及右下叶肺组织,采用Taqman探针法检测CCL2、CCL7和CCR2 mRNA的表达水平。结果 (1)淡水溺死组大鼠解剖征象及肺组织HE染色结果符合溺死的特征,疾病对照组大鼠心肌组织HE染色结果符合心肌梗死的病理改变;(2)各组大鼠肺组织中CCL2、CCL7和CCR2mRNA的表达水平:淡水溺死组CCL2和CCL7 mRNA的表达水平显著高于空白对照组及淡水应激组(P<0.05),且淡水溺死组CCR2 mRNA表达水平高于淡水应激组(P<0.05);与疾病对照组比较,无统计学差异(P>0.05)。(3)各组大鼠血清中CCL2、CCL7和CCR2 mRNA表达水平:淡水溺死组血清中CCL2、CCL7和CCR2 mRNA的表达水平均高于对照组水平(P<0.05)。结论采用Taqman探针法检测肺组织及血清中CCL2、CCL7和CCR2 mRNA的表达水平,对溺死的诊断有一定的辅助作用。Objective We detected the expression of CCL2, CCL7 and CCR2 mRNA in lung tissue and serum of rats after freshwater drowning to evaluate the potential applications in identifying the drowning deceased. Methods Twenty four rats were randomly divided into four groups: the freshwater drowning group ( n = 6) , the freshwater stress group ( n = 6) , the disease control group ( n = 6) , the blank control group ( n = 6). Part of left ventricular muscle tissue and right lower lobe lung tissue from each rat were collected for the Hematoxylin and Eosin (HE) staining and meanwhile, A appropriate amount of the right ventricle serum and interior lobe of right lung tissue to detect the expression of CCL2, CCL7 and CCR2 mRNA by using TaqMan Probe. Results In the lung tissue, the expression of CCL2 and CCL7 mRNA of the freshwater drowning group was higher ( P 〈 0. 05 ) comparing with the blank control group ; moreover, the CCL2, CCL7 and CCR2 mRNA levels were also higher than near-drowning stress group. However, there was no statistically significant differences when compared with the disease control group. In the serum, the expression of CCL2, CCL7 and CCR2 mRNA significantly increased in the drowning group comparing with the other control groups( P 〈 0. 05 ). Conclusion Detecting the expression of CCL2, CCL7 and CCR2 mRNA in serum and lung tissue will contribute to identifying the drowning deseased forensic medicine application.
