紫锥菊颗粒对大鼠的亚急性毒性试验

摘　　要：为了探讨紫锥菊颗粒对大鼠的毒性作用,评价其临床用药的安全性,将80只Wistar大鼠随机均分为4组,每组20只。每组分别以2 000/mg/kg、1 000 mg/kg、500 mg/kg和0 mg/kg经口染毒,1次/d,连续染毒30 d,每日观察并记录各组大鼠的一般行为表现,每周测量一次大鼠体质量并调整染毒剂量。试验结束后采集血样,测定各组大鼠的血液学和血清生化参数,扑杀所有存活大鼠,进行眼观检查,并对肝、肾、脾、肺等脏器进行组织病理学检查。结果显示,试验期间各剂量组大鼠均未死亡,且生长发育良好,血液学和血清生化参数中仅高剂量组葡萄糖、天门冬氨酸氨基转移酶及中剂量组中肌酐与对照组相比有显著差异（P〈0.05）,但均在正常范围内,且无剂量相关性。各剂量组大鼠的眼观和组织病理学检查均未发现异常病变。结果表明,紫锥菊颗粒长期重复应用无亚急性毒性。To study the subacute toxicity of Echinacea purpurea granules on rats and evaluate its clinical safety,eighty Wistar rats were randomly divided into 4 groups with 20 rats in each groups and orally given Echinacea purpurea granules at a dose of2 000 mg/kg,1 000 mg/kg,500 mg/kg and 0 mg/kg once daily for 30 days. The general behavior was observed and recorded every day,the weight was measured every week to adjust the oral dose accordingly. After the test,blood samples were collected to detect physiological and biochemical parameters,all the rats were slaughtered to macrography and histopathologic examination of liver,kidney,spleen and lung. The results showed that all the rats were survived and grew well. For hematology and blood biochemical parameters,only GLU and AST in high group and CRE in middle dose group were significantly different from the control group（0.05P0.01）,but numerical value within normal limits and was not dose-dependent. All rats were not found abnormal lesion through macrography and histopathologic examination. The results suggested that Echinacea purpurea granules has no subacute toxicity.
