氧化白藜芦醇对小鼠肝癌淋巴结转移的抑制作用

摘　　要：目的考察氧化白藜芦醇对小鼠肝癌淋巴结转移的抑制作用。方法 70只小鼠随机均分为正常对照组、模型组、阿霉素组(5 mg/kg)、白藜芦醇组(40 mg/kg)、氧化白藜芦醇低、中、高剂量组(20、40、60 mg/kg),各组腹腔注射给药21 d。然后,测量小鼠体质量及肿瘤、淋巴结体积和质量,计算免疫器官指数,检测淋巴结转移情况及MMP-9、VEGF-C mRNA表达。结果与模型组比较,氧化白藜芦醇组小鼠体质量、胸腺指数、脾脏指数无显著差异(P>0.05);肿瘤、淋巴结体积和质量明显减少(P<0.05,P<0.01),并呈浓度依赖性;淋巴结转移更少;MMP-9、VEGF-C mRNA表达明显降低(P<0.05,P<0.01)。结论氧化白藜芦醇可有效抑制小鼠肝癌淋巴结转移,并对免疫器官的影响较小。AIM To investigate the inhibitory effects of oxyresveratrol on lymph node metastasis of mouse hepatocellular carcinoma. METHODS Seventy mice randomly and equally divided into normal control group,model group,adriamycin group（ 5 mg/kg）,resveratrol group（ 40 mg/kg）,and high,medium,and low dose oxyresveratrol groups（ 60,40,20 mg/kg）,were given intraperitoneal injection administration,respectively,for measurements of the body weights of mice,the volumes and weights of tumors and lymph nodes,calculation of immune organ indices,detection of the lymph node metastasis condition and MMP-9,VEGF-C mRNA expressions.RESULTS The oxyresveratrol groups were observed with significantly reduced volumes,weights of tumors and lymph nodes（ P〈0. 05, P〈0. 01） in a concentration-dependent manner,less lymph node metastasis,and significantly decreased MMP-9,VEGF-C mRNA expressions（ P〈0. 05,P〈0. 01）,but no significant differences in mice body weights,and indices for thymus and spleen（ P〈0. 05） as compared with the model group.CONCLUSION Oxyresveratrol can effectively inhibit the lymph node metastasis in hepatocellular carcinoma mice with little impact on immune organs.
