摘 要:目的建立一种快速简便地测定荜茇药材中荜茇酰胺含量的方法,并用此方法测定不同产地及不同药用部位荜茇药材中荜茇酰胺含量。方法以荜茇药材中荜茇酰胺为指标成分,采用迪马Diamond C18(5μm 250×4.6 mm)色谱柱,流动相为甲醇-水(35∶65),流速1 m L·min-1,柱温30℃,进样量20μL,检测波长254 nm,测定荜茇药材中荜茇酰胺含量,并用该方法测定海南、广西、四川等产地荜茇药材中荜茇酰胺的含量,并比较其差异。结果荜茇药材中的荜茇酰胺含量在0.0414~0.3726 mg·m L-1与峰面积线性关系良好(r=0.9999),平均回收率为98.58%,RSD=1.03。不同产地荜茇药材中,广西壮族自治区产地的荜茇中荜茇酰胺含量最高,荜茇茎中荜茇酰胺的含量明显低于荜茇果实。结论该方法建立了一种快速、准确、经济的测定荜茇药材中荜茇酰胺含量的方法,为荜茇药材的质量控制与品质评价奠定了基础,不同产地及不同药用部位的荜茇中荜茇酰胺含量存在差异。Objective To establish a quick and easy deter mination of Piper Longum amide content in Piper Longum L. drug and compare their differences from different origins and different medicinal parts, and to lay the foundation for the quality control of Chinese medicinal materials. Methods The quantitative determination of Piper Longum amide was performed by HPLC with a Diamond C18 chromatographic column(250 mm × 4.6 mm, 5 μm) and mobile phase composed of methanol-H2 O(35: 65) at the flow rate of 1.0 m L·min-1. The column temperature was 30℃ and the UV detection wavelength was 254 nm. The Injection Volume was 20μL. Results The linear relationship was good in the range of 0.0414-0.3726 mg m L-1(r=0.9999). And the average recovery rate was 98.58%(n=6) with the RSD of 1.03%. The content of Piper Longum amide in Piper Longum drug from different area and different medicinal parts is significant different. The content of Piper Longum amide from Guangxi Zhuang Autonomous Region origin was the highest and the content in the stems of Piper Longum amide was significantly lower than the fruit. Conclusion A rapid,accurate and economical method was established to determined Piper Longum amide in piper longum drug. It laid a foundation for evaluating quality of Piper Longum drug drug.
