脑胶质瘤组织中lncRNA-LINC01116的表达变化及其意义

摘　　要：目的观察脑胶质瘤组织中长链非编码RNA(lncRNA)-LINC01116的表达变化,并探讨其意义。方法脑胶质瘤组织95例份,正常脑组织21例份,采用实时荧光定量PCR法检测lncRNA-LINC01116;分析lncRNALINC01116表达与脑胶质瘤临床病理特征及患者预后的关系。结果脑胶质瘤、正常组织lncRNA-LINC01116的相对表达量分别为0.665±0.678、0.103±0.147,两者比较,P<0.05。lncRNA-LINC01116仅与脑胶质瘤患者临床病理分级有关(P<0.05),而与患者年龄、性别、是否发生脑积水、病理类型、肿瘤直径、术前KPS评分、侵犯脑叶数目无关(P均>0.05)。按照lncRNA-LINC01116相对表达量的平均数0.665为界,将95例患者分为低表达者41例和高表达者54例,低表达者生存率为73.17%(30/41)、生存时间为(100.30±6.91)个月,高表达者生存率为31.48%(17/54)、生存时间为(43.90±5.92)个月,两者比较,P均<0.05;低表达者复发率为31.71%(13/41),高表达者复发率为74.07%(40/54),两者比较,P<0.05。结论脑胶质瘤组织中lncRNA-LINC01116表达升高,其表达与临床病理分级及患者预后相关;lncRNA-LINC01116有望成为脑胶质瘤的病理诊断、预后的肿瘤标记物。Objective To observe the expression changes of long chain non-coding RNA（ lncRNA）-LINC01116 in the glioma tissues and to explore its significance. Methods The lncRNA-LINC01116 was detected in 95 cases of glioma tissues and 21 cases of normal brain tissues by real-time fluorescence quantitative PCR. The relationship between lncRNALINC01116 expression and clinicopathological features and prognosis were analyzed. Results The relative expression of lncRNA-LINC01116 in the glioma and normal tissues was 0. 665 ± 0. 678 and 0. 103 ± 0. 147,respectively,and significant difference was found（ P〈0. 05）. lncRNA-LINC01116 was related to the clinicopathological grade of glioma（ P〈0. 05）,and was not related to the age,sex,hydrocephalus,pathological type,tumor diameter,preoperative KPS score,or the number of cerebral lobe involved（ all P〉0. 05）. According to the average number of relative expression of lncRNALINC01116（ 0. 665）,95 patients were divided into the low expression group（ n = 41） and the high expression group（ n =54）; the survival rate of the low expression group was 73. 17%（ 30/41）,and the survival time was（ 100. 30 ± 6. 91）months,and the survival rate of the high expression group was 31. 48%（ 17/54）,and the survival time was（ 43. 90 ±5. 92） months; significant difference was found between these two groups（ both P〈0. 05）. The recurrence rate of the low expression group was 31. 71%（ 13/41）,and the recurrence rate of high expression group was 74. 07%（ 40/54）,and significant difference was found,P〈0. 05. Conclusions The expression of lncRNA-LINC01116 in the glioma tissues is increased,which is closely related to the clinical grades and prognosis of patients. lncRNA-LINC01116 is expected to be a pathological diagnosis and prognostic tumor marker for glioma.
