安徽省西瓜枯萎病菌遗传多样性ISSR-PCR分析

摘  要:以42株来自安徽省不同地区的西瓜枯萎病菌为试材,采用Inter-simple Sequence Repeats(ISSR)技术对其群体的遗传多样性进行分析,探究病菌的遗传分化与地理来源之间的相关性.结果表明,用筛选出的11个随机引物对西瓜枯萎病菌菌株进行ISSR-PCR扩增,共获得129个ISSR条带,其中多态性条带为94个,占72.9%,表明安徽省西瓜枯萎病菌存在较丰富的遗传多样性,菌株间的遗传相似系数为0.71~0.97.UPGMA聚类分析显示,供试菌株可区分为8个聚类组,菌株的遗传谱系与地理来源无相关性.In order to probe into the relationship between genetic diversity and geographical origin among isolates of Fusarium oxysporumf.sp.neviumfrom Anhui Province in eastern China,inter-simple sequence repeat-polymerase chain reaction(ISSR-PCR)was conducted to investigate the genetic diversity of 42 isolates of F.oxysporum f.sp.nevium.The result showed that 11 random primers which produced reproducible fragments were screened,generated a total of 129 ISSR fragments,of which 72.9%(94)were polymorphic,revealing high polymorphism among the isolates.Genetic similarity coefficients among all of the isolates ranged from 0.71 to 0.97.Cluster analysis using the unweighted pair-group method with arithmetic averages(UPGMA)indicated that the Anhui isolates were divided into eight groups according to the DNA fingerprints,and that no correlation between ISSR group and geographical origin.
