摘  要:目的比较两种不同类型无细胞百日咳疫苗免疫小鼠后诱导体液免疫和细胞免疫应答的特点。方法将BALB/c小鼠分为3组(分别纯化Pa组、共纯化Pa组、PBS组),分别用稀释后的两种不同工艺生产的无细胞百日咳疫苗及PBS皮下接种,接种量0.5 m L/只,于第4周加强免疫,剂量与初免相同。于免疫后第1周、第4周和第5周取血,分离血清用于检测小鼠抗百日咳抗体Ig G水平(抗-PT和抗-FHA);同时于加强免疫后第7天,分离小鼠脾淋巴细胞,经刺激后,取其培养上清,用流式细胞微球芯片捕获法(cytometric bead array,CBA法)测定脾淋巴细胞经体外刺激后所产生的Th1、Th2和Th17型细胞因子的含量。结果对两种类型无细胞百日咳疫苗组检测的结果进行比较,在免后第4周,分别纯化Pa组和共纯化Pa组的抗-PT和抗-FHA Ig G水平差异无统计学意义(P>0.05);但在免后第5周,分别纯化Pa组的抗-PT和抗-FHA Ig G水平均高于共纯化Pa组,差异均具有统计学意义(P<0.05)。经PT刺激后诱导产生的细胞因子,分别纯化Pa组的IL-4含量高于共纯化Pa组,差异具有统计学意义(P<0.05);分别纯化Pa组和共纯化Pa组的其他细胞因子差异均无统计学意义(P>0.05)。结论两种类型的无细胞百日咳疫苗免疫小鼠后均能诱导产生体液免疫应答和细胞免疫应答。Objective To compare the characteristics of humoral immune and cellular immune responses induced in mice by two different types of pertussis vaccines. Methods Diluting the pertussis vaccines produced by two different processes,three groups of tested mice were given subcutaneous inoculation,0.5 m L per mouse,and immunization was strengthened at the fourth week and the dose was the same as the first immunization. After the first,fourth and fifth weeks of immunization,blood was taken and serum was separated for the detection of anti-pertussis antibodies( anti-PT and anti-FHA). At the same time,on the seventh day after the booster immunization,the mice spleen lymphocytes were isolated,stimulated,and cultured. Determination of Th1/Th2/Th17 cytokines was carried out by Cytometric Bead Array( CBA) from splenic lymphocytes. Results There was no a statistical difference( P〉0.05) in the levels of anti-PT and anti-FHA between the two groups in the fourth week after immunization; however,there were statistical differences( P〈0.05) in the levels of anti-PT and anti-FHA in the fifth week,the level in groups by using separated purification higher than that of groups by co-purification. In terms of cytokines,there was a statistical difference( P〈0.05) in the concentration of IL-4 between the two groups after PT stimulation. There were no statistical differences( P〉0. 05)between other cytokines. Conclusion The humoral immune response and cellular immune response were developed by immunization of both types of pertussis vaccines in mice.
