摘 要:目的研究丹参注射液对糖尿病大鼠海马组织中葡萄糖调节蛋白-78(Grp-78)以及生长停滞和DNA损伤损伤基因153(CHOP/GAD153)表达的影响。方法将30只SD大鼠按照随机数字表分为对照组、糖尿病组和丹参注射液组,每组各10只。糖尿病组、丹参注射液治疗组的大鼠采用单次腹腔注射链脲佐菌素65mg/kg诱导糖尿病模型。48h后,丹参注射液治疗组每天腹腔注射丹参注射液1ml/kg,对照组和糖尿病组腹腔注射等量0.9%氯化钠溶液。各组均连续注射6周,随后测定各组大鼠体重和血糖。采用水迷宫检测依赖海马的学习记忆能力,超氧化物歧化酶(SOD)和丙二醛(MDA)试剂盒检测海马组织匀浆中SOD活性以及MDA含量,免疫印迹检测海马组织中cleaved caspase-3蛋白表达,免疫组化检测海马组织中Grp-78和CHOP蛋白表达。结果与对照组相比,糖尿病组大鼠体重减轻,血糖升高(P<0.01),水迷宫检测中的逃避潜伏期明显变长(P<0.01),穿越平台次数显著增多(P<0.01),同时,海马组织中SOD活性降低(P<0.01),MDA含量、Grp-78、CHOP和cleaved caspase-3表达均升高(均P<0.01)。而与糖尿病组相比,丹参注射液治组大鼠体重增加,逃避潜伏期明显减短,穿越平台次数显著减少,SOD活性明显提高(P<0.01),MDA含量、Grp-78、CHOP以及cleaved caspase-3表达均减少(均P<0.01)。结论丹参注射液能够抑制糖尿病大鼠海马组织中内质网应激的激活,从而减少神经元的凋亡,提高大鼠的学习记忆能力。Objective To investigate the effect of Danshen(Salvia miltiorrhiza)diabetic injection on and the expression Spragueof Grp-78rats(Glucose regulated protein-78)and CHOP(GAD153)in hippocampus of rats.Methods Thirty Dawley were randomly divided(STZ)into(65 three mg/kg)Danshen groups was with 10 in in rats each of group:diabetes control group group,and diabetes group Danshen to injection diabetes group.model.q.d.Streptozotocin After injected Danshen injection in group induce 48h of STZ injection,equal injection(1ml/kg)was administered intraperitoneally rats of Danshen injection weeks,group,for6 weeks,and while volume were of normal saline all was injected Morris in rats of maze control and used diabetes to groups.the After 6 the body weight ability blood glucose measured in groups.water was detect hippocampus-dependent(MDA)of learning were and memory.The activity of superoxide dismutase(SOD)and the contents of malondialdehyde in hippocampus measured.The expression of cleaved caspase-3 was detected The by immunoblotting and the and expression the levels glucose of Grpwas 78 and CHOP the were escape detected latency by and immunohistochemistry.the Results in body weight was decreased blood elevated,average frequency finding the platform in diabetes group were significantly increased when compared CHOP with control group caspase-(P〈0.01).Meanwhile,the reduction of SOD activity and the increased expression ofMDA,Grp-78,and cleaved 3 were observed in hippocampus of diabetic rats(P〈0.01).However,Danshen injection increased body weight and the reduced escape the expression levels average of MDA,Grp-78,CHOP and cleaved caspase-3 with a increased SOD activity.Danshen Meanwhile,injection latency neuronal and the frequency improve in finding ability the platform were also decreased.via Conclusion may reduce apoptosis and the of learning and memory suppressing the activation of endoplasmic reticulum stress.
