摘 要:杂交育种过程中的异地授粉和花期不遇问题一直是困扰着科研工作者的难题。因此,亟需探究出一种能长时间保存花粉且能较好地保留花粉活性的方法。以光果姜、珊瑚姜和红球姜3种姜属植物花粉为试验材料,筛选花粉活性最佳检测方法、检测花粉常温下的活性变化并探讨花粉最佳贮藏条件,以解决杂交授粉中异地授粉和花期不遇的问题。结果表明:MTT染色法能快速检测3种姜属植物花粉活性;在自然状态下,3种姜属植物在刚开花时花粉活性较高,花粉活性随放置时间的延长而降低,光果姜花粉常温下的放置时间较其余2种花粉时间长;光果姜花粉和红球姜花粉的最佳贮藏方法为-80℃不干燥,珊瑚姜花粉最佳贮藏方法为-80℃干燥,最佳解冻方法均为直接取出。The problem of ex situ pollination and different flowering time at cross breeding stage has always been an troublesome issue for the breeders. Therefore, it is very necessary to develop a method to store pollen for a longer time and to retain well the pollen viability. Pollen of Zingiber nudicarpum, Z.corallinum and Z. zerumbet were stored under various conditions to select their optimum storage conditions, and their viability was detected to observe its change at the ambient temperature to select an optimum detection method for pollen. The results showed that the pollen viability of the three species of Zingiber was tested accurately by MTT staining. Under the natural condition, the pollen viability of these three species of Zingiber was high at the initial flowering stage and decreased with time. The pollen of Z.nudicarpum were stored for a longer time than that of the other two Zingiber species under the room temperature. The best storage condition for pollen of Z. nudicarpum and Z. zerumbet was to store at-80℃,while the pollen of Z. corallinum were stored dry at-80℃. The best thawing method is to take the pollen out directly from the storage.
