摘  要:目的观察开智灵组方对改善记忆的影响及其作用机制。方法小鼠随机分为六组,分别为正常对照组、开智灵组方低剂量组、中、高剂量组、核桃酶解提取物组(66.67 mg/kg)、牛磺酸组(33.33 mg/kg),实验组小鼠每天灌胃给药0.2 m L/10 g,每日一次,30 d后通过避暗实验、水迷宫实验测试学习成绩,并进行重复性实验和消退实验。在避暗实验重复性实验结束后,试剂盒测定小鼠海马中超氧化物歧化酶(SOD)、丙二醛(MDA)含量;在水迷宫实验重复性实验中,Western blot测定小鼠海马中脑源性神经营养因子(BDNF)、酪氨酸激酶B(Trk B)蛋白含量表,Real-time PCR测定m RNA含量表达。结果开智灵组方在小鼠避暗实验、小鼠水迷宫实验前后两次结果均为阳性。避暗实验重复性实验中各给药组小鼠海马组织中SOD含量有升高趋势、MDA含量有降低趋势,但差异无统计学意义(P>0.05)。水迷宫实验重复性实验中各给药组小鼠海马组织中BDNF、Trk B的m RNA和蛋白表达量均上调,差异有统计学意义(P<0.05)。结论开智灵组方对小鼠具有良好的改善记忆的作用,其机制可能与调节脑内BDNF及Trk B表达有关。Objective To investigate the effect of kaizhiling formulation on learning and memory ability and explore itsmechanism. Methods Mice were randomly divided into six groups: control group, kaizhiling group( low, middle, high dose),enzymatic hydrolysate of walnut group(66.67 mg/kg), taurine group(33.33 mg/kg). Mice were given intragastricadministration of drugs 0.2 m L/10 g once a day for consecutive 30 days. Passive avoidance task and Morris water maze testwere performed to evaluate ability of learning and memory. Repeated experiment and regression experiment were alsooperated. At the end of the experiment, malondialdehyde(MDA), superoxide dismutase(SOD) content of hippocampus weredetermined by commercial kits; the protein expression of brain derived neurotrophic factor(BDNF) and tyrosine kinase B(Trk B) were analyzed by Western blot. The m RNA expression of BDNF and Trk B were analyzed by Real-time PCR.Results Kaizhiling formulation was positive in both the Passive avoidance task and Morris water maze test. The content ofSOD in the hippocampal tissues of each drug group was increased and the MDA content decreased, but the difference wasnot statistically significant(P〉0.05). The m RNA and protein of BDNF and Trk B was overexpressed in the hippocampaltissues of each drug group, and the differences were statistically significant(P〈0.05). Conclusion Kaizhiling formulationenhance the learning and memory ability in mice. The mechanism may be related to the regulation of BDNF and Trk Bexpression in the hippocampus.
