摘 要:目的探讨参麦注射液对内毒素诱导心肌细胞凋亡的影响。方法原代培养乳鼠心肌细胞,分为对照组,内毒素(LPS)处理组,参麦注射液(SMI)+内毒素(LPS)处理组。MTT法检测心肌细胞存活率;流式细胞仪检测心肌细胞凋亡;RT-PCR检测心肌细胞Wnt2、β-catenin m RNA的表达;Western blot检测心肌细胞内Wnt2、β-catenin蛋白表达。结果0.8 g/L与1 g/L SMI组心肌细胞存活率高于LPS组;0.8 g/L与1 g/L SMI组心肌细胞凋亡率低于LPS组。SMI+LPS组中Wnt2、β-catenin m RNA的表达随SMI剂量的增加而降低,0.8 g/L SMI组中β-catenin m RNA表达低于LPS组,1g/L SMI组中Wnt2、β-catenin m RNA表达低于LPS组;SMI+LPS组中Wnt2、β-catenin蛋白的表达随SMI剂量的增加而降低,0.8 g/L SMI组中β-catenin蛋白表达低于LPS组,1g/L SMI组中Wnt2、β-catenin蛋白表达低于LPS组,差异有统计学意义(P<0.05)。结论参麦注射液对内毒素诱导的乳鼠心肌细胞凋亡具有明显的抑制作用,其机制可能与抑制Wnt2、β-catenin的表达有关。Objective To investigate the effects of Shenmai injection on cardiomyocyte apoptosis induced by endotoxin.Methods Primary cultured neonatal rat cardiomyocytes were divided into control group, endotoxin(LPS) treatment group,Shenmai injection(SMI) + endotoxin(LPS) treatment group.The survival rate of cardiomyocytes was detected by MTT; apoptosis of cardiomyocytes was detected by flow cytometry; expression of Wnt2 and beta-catenin m RNA in cardiac myocytes was detected by RT-PCR; and the expression of Wnt2 and beta-catenin protein in cardiac myocytes was detected by Western blot. Results Cardiomyocyte survival rate of 0.8 g/L and 1 g/L SMI group was higher than that of LPS group.The apoptosis rate of myocardial cells in 0.8 g/L and 1 g/L SMI group was lower than that in LPS group. In the SMI+LPS group, the expression of Wnt2 servated-catenin m RNA decreased with the increase of SMI dose. In the 0.8 g/L SMI group, the expression of-catenin m RNA was lower than that of the LPS group, and in the 1 g/L SMI group, the expression of Wnt2 minus catenin m RNA was lower than that of the LPS group.In the SMI +LPS group, the expression of Wnt2 isocatenin protein decreased with the increase of SMI dose. In the 0.8 g/L SMI group, the expression of-catenin protein was lower than that in the LPS group, and in the 1 g/L SMI group, the expression of Wnt2-catenin protein was lower than that in the LPS group.The difference was statistically significant(P〈0.05). Conclusion Shenmai injection has a significant inhibitory effect on LPS induced apoptosis in neonatal rat cardiac myocytes, and its mechanism may be related to the inhibition of the expression of Wnt2 and beta-catenin.
