胃肠舒片醇提物不同剂量对大鼠胃肠动力及胃肠激素的影响

摘　　要：目的通过胃肠舒片醇提物不同剂量对大鼠胃肠动力和胃肠激素影响的研究,探讨其临床最佳用药剂量所达到的最佳疗效。方法选SD大鼠随机分4组,每组6只,即空白组,胃肠舒小剂量组、中剂量组和高剂量组。空白组给蒸馏水,胃肠舒三个剂量组分别给予胃肠舒小剂量灌胃液、中剂量灌胃液、高剂量灌胃液,各组灌胃用药7天后,以葡聚糖蓝(BD)2000为标记物,观察各组胃排空率和小肠推进率,检测各组大鼠血清胃泌素(GAS)与胃动素(MTL)的含量,并进行对比分析。结果胃肠舒小剂量和高剂量组促进大鼠胃排空率显著优于空白组(P<0.01或P<0.05);胃肠舒小、中、高剂量组促进大鼠小肠推进率均显著优于空白组(P<0.01,P<0.05);胃肠舒小剂量组增强大鼠血清胃动素(MTL)含量显著优于空白组(P<0.01)。随胃肠舒片剂量增加反而有对促进胃肠动力和胃肠激素分泌作用减弱趋势,但无统计学意义。结论小剂量胃肠舒片即可达到促进胃肠动力和增加MTL的最佳疗效。Objective To investigate the best curative effect of the optimum dosage in clinic, effects of different doses of ethanol extract of Weichangshu pills on gastrointestinal motility and gastrointestinal hormones in rats were studied. Methods SD rats were randomly divided into 4 groups, the blank group, Weichangshu pills low dose group, Weichangshu pills middle dose group and？ Weichangshu pills high dose group,6 rats in each group. The blank group was given distilled water, the other three groups were given low, medium, and high doses of Weichangshu pills. Rats in each group were given intragastric administration for seven days, the gastric emptying rate and intestinal propulsive rate were observed with dextran blue （BD） 2000 as the marker, and the levels of serum gastrin （GAS） and motilin （MTL） in each group were detected and compared. Results The gastric emptying rate of rats in the low and high dose group was significantly better than that in the blank group （ P 〈 0.01 or P 〈 0.05 ）. The low, middle and high dose groups promoted the intestinal propulsive rate of rats which were significantly better than that of the blank group （P 〈 0.01 or, P 〈 0.05）. The level of serum motilin （MTL） in rats with the low group was significantly higher than that in the blank group （P 〈0.01 ）. With the increase of the dosage of Weichangshu pills, the gastrointestinal motility and secretion of gastrointestinal hormones decreased, but there was no statistical significance. Conclusion Low dose of Weichangshu pills can a- chieve the best effect of promoting gastrointestinal motility and increasing MTL.
