摘  要:目的:观察持续光照对大鼠卵巢、子宫的影响及中药干预作用。方法:185只动情周期正常的2月龄性成熟雌性SD大鼠,随机选取60只入对照组,予正常光照。剩余大鼠给予光照强度为(300±20)lux的24h/d持续光照的条件,持续50d,造成昼日节律紊乱。将成模大鼠随机分为模型组(n=50)和中药组(n=50)。中药组给予滋阴补阳序贯中药治疗,其余两组给予等体积生理盐水灌胃,观察各纽大鼠动情周期的变化。20d后,测定大鼠褪黑素水平,留取卵巢、子宫,行HE染色后进行形态学评估。结果:对照组大鼠呈现规律的动情周期变化,模型组大鼠表现为几乎持续的阴道角化细胞,而中药组大鼠可间或出现动情前期、动情后期、动情间期。对照组褪黑素呈现昼低夜高的近日节律变化,模型组与中药组丧失了近日节律,在夜间显著低于对照组,在白间高于对照组;中药组褪黑素22:00显著高于模型组。模型组大鼠卵巢湿重低于对照组和中药组。卵巢形态学比较,对照组可见较多的发育中的卵泡和黄体,而模型组表现为成熟卵泡和黄体减少,囊状扩张的卵泡和退化黄体增多。中药组成熟卵泡和黄体高于模型组,囊肿卵泡和退化黄体低于模型组,囊肿卵泡高于对照组。子宫形态学比较,模型组腺上皮增生活跃,腺体发达,形态评分显著高于对照组和中药组。结论:持续光照可导致大鼠动情周期紊乱,卵巢、子宫损伤,中药可以诱导周期重建,对于卵巢和子宫具有保护作用,机制可能与中药提高褪黑素水平有关。Objective : To observe the effect of continuous light on ovary and uterus of rats and intervention effect of tra- ditional Chinese medicine. Methods:A total of 182 sexual maturity female SD rats in the age of 2 months with norrmal es- trous cycle were enrolled. Sixty rats were selected randomly into control group and were given normal light. The other rats were given ( 300± 20) lux light intensity for 24 h/d, lasting for 50 days to induce circadian rhythm disorder. At last, we got 50 rats enrolled into model group and 50 rats enrolled into herbal group. The herbal group rats were given therapy of nourishing Yin and tonifying Yang herbal while the other two groups were given physiological saline. Twenty days later, blood was taken out and melatonin was tested. The ovary and uterus morphology was observed after HE stained. Results: Rats in control group showed regular estrous cycles. Rats in model group showed persistent cuticulated epithelium. Rats in herbal group showed preoestrus metestrum and anestrus sometimes. Melatonin in control group exhibited circadian rhythm, high at night but low in the day, while model group and herbal group displayed disordered circadian rhythm. Me- latonin in model group and herbal group were decreased compared with that of the control group during night and higher during day. Melatonin in herbal group was higher than that of the model group at 22 : 00. Ovary wet weight in model group was decreased compared with that of the control group and herbal group. Ovary morphology:development follicles and corpus luteum in control group were observed. There were decreased developed follicles and corpus luteum, increased cystic expansion follicles and degraded corpus luteum in model group compared with those of the control group. Developedfollicles and corpus luteum in herbal group were more than those in model group, while cystic expansion follicles and de- graded corpus luteum were less than those of the model group.
