摘 要:[目的]筛选出高生根率培养基配方和适宜的光照条件。[方法]以金花茶组培壮芽为材料,研究培养基养分含量、生长素种类组合和浓度配比对金花茶组培芽生根的影响,筛选适宜生根的光照条件。[结果]J、1/2 J、1/3 J和2/3 J基本培养基对金花茶组培芽生根率的影响存在显著差异,大小排序为2/3 J>1/2 J>1/3 J>J;不同浓度的IBA、IAA、NAA组合对金花茶组培芽生根率影响差异显著,正交极差分析和方差分析结果显示,IAA起主导作用,IBA起辅助作用;在2/3J培养基中添加不同浓度Ca2+,结果表明培养基Ca2+浓度以2/3J的79.20 mg/L基础上添加19.60 mg/L处理对金花茶组培芽生根培养比较适宜。[结论]最佳诱导金花茶组培芽生根培养基配方为2/3J+Ca2+16.90 mg/L[100 mg/L Ca(NO3)2·4H2O]+2.0 mg/L IBA+5.0 mg/L IAA+0.3 mg/L NAA。[ Objective ] The formulation of hi/h rootin/ rate culture medium and appropriate li/htin/ conditions were screened out. [ Method ]Camellia nittidissima Chi tissue culture shoot was chosen as study materials in this experiment. This study invemedium,type and concentration of auxin,concentration of calcium and li/htin/ condition. [Result] There was si/nificant difference in rootin/rate amon/ basic mediums of J,1/2 J,1/3 J, 2 3 J,in which the 2/3 J medium was the best,a d the followin/ was that in 1 2 J,1/3 J and J. Rootin/ rate did differ si/nificantly amon/ combinations of concentrations of auxins includin/ IBA, IAA and NAA, in which shows that IiVA plays a leadin/ role and IBA plays a subsidiary role. The concentration of calcium addin/ 19. 60 m/L fered a better /rowth condition to shoot of tissue culture. [ Conclusion] The optimal rootin/ medium for C. nittidissima tissue culture shoot was2/3J+Ca2+ 16. 90 m/L [ 100 m/L Ca(W〇) 2 ? 4H20 ] + 2 . 0 m/L IBA+5. 0 m /L IAA+0. 3 m /L NAA.
