摘 要:目的:米非司酮作为目前公认的紧急避孕药物,为一种选择性孕激素受体调节剂,广泛应用于临床。然而,米非司酮对精子的作用研究甚少。本研究探索米非司酮对人精子功能的体外生物学效应。方法:精子在获能后孵育在含0.04μM、0.4μM、4μM、40μM的米非司酮培养液中1h。检测精子活力、活率、DNA完整性、顶体反应、精子超活化、精子内游离钙离子浓度。结果:经过使用不同浓度的米非司酮体外处理精子,检测精子的活率、活力、形态、DNA完整性,对照组与实验组差异无统计学意义(P>0.05),当米非司酮的浓度大于0.4μM时显著地降低了精子超活化的比率(P<0.01)。进一步检测处理后的精子顶体反应率和精子内钙离子浓度发现,在米非司酮浓度大于0.4μM时,抑制了顶体反应并且降低了精子内钙离子浓度。结论:米非司酮在体外的杀精机制是通过抑制顶体反应、超活化以及降低钙离子浓度来实现的。Objective: Mifepristone,as an accepted emergency contraceptive pill,is a selective progesterone receptor modulator and widely used in clinic. However,the effect of mifepristone on sperm is poorly studied. In this study,the effects of mifepristone on human sperm in vitro was investigated. Methods: Spermatozoa were incubated in the culture medium containing 0. 04μM,0. 4μM,4μM and 40μM of mifepristone for an hour after capacitation. The sperm viability,motility,DNA integrity,acrosome reaction,sperm hyperactivation,and intracellular free calcium concentration were measured. Results: After treatment,differences in the viability,motility,morphology and the integrity of DNA were not significant between control group and treatment group( P 〉 0. 05). When mifepristone concentration was greater than 0. 4μM,the ratio of sperm hyperactivation was significantly reduced( P 〈 0. 01). According to further examination of the acrosome reaction and sperm intracellular calcium concentration,when mifepristone concentrations was greater than 0. 4μM,the acrosome reaction was significantly inhibited and the calciumion concentration in spermatozoa decreased( P 〈 0. 01). Conclusions: The mechanism of mifepristone in vitro is achieved by inhibiting acrosome reaction,hyperactivation and decreasing calcium concentration.
