摘 要:目的探讨通心络对肥大细胞介导的大鼠心肌缺血再灌注损伤中β-氨基己糖苷酶(β-hexosaminidase,β-Hex)、类糜蛋白酶及蛋白激酶C(protein kinase C,PKC)表达的影响,进而探寻通心络对受损心肌的保护作用及相关机制。方法将36只健康成年雄性SD大鼠随机分成假手术组、模型组、通心络组,每组12只。通过可逆性左冠状动脉前降支结扎法建立心肌缺血再灌注模型,大鼠心肌缺血1小时,再灌注2小时;假手术组仅穿线不结扎。通心络组大鼠予通心络灌胃,假手术组和模型组给予等量的生理盐水灌胃。1周后,采用RT-PCR法检测左心室心肌组织PKCε和PKCδmRNA的表达;Western blot法检测PKCα的表达水平。Elisa法分别检测心肌组织β-氨基己糖苷酶和类糜蛋白酶的水平。结果通心络组大鼠β-氨基己糖苷酶、类糜蛋白酶及PKC表达水平较模型组均明显降低(P<0.01或P<0.05)。结论通心络可以降低β-氨基己糖苷酶、类糜蛋白酶及PKC表达水平,抑制肥大细胞脱颗粒,减轻受损心肌的炎症反应,进而保护心肌组织。Objective To investigate the effects of Tongxinluo on the expression of β- hexosaminidase, chymase and PKC in rats with myocardial ischemia reperthsion injury mediated by mast cells, and to explore the protective eftect of Tongxinluo capsule on mocardial injury and the related mechanism. Methods 36 healthy adult male SD rats were randomly divided into sham operation group, model group, Tongxinluo group. Myocardial ischemia reperthsion model was established by ligation of the left anterior descending coronary artery, Rat myocardial ischemia 1 h, reperthsion 2 h; while the sham group was punctured without ligation. The rats in Tongxinluo group were ted with Tongxinluo suspension. And the model group and the sham group were ted with saline. A week later, the expressions of PKCε and PKCδ mRNA in left ventricular myocardium were detected by RT-PCR, and the expression of PKC was checked by Western blot. Elisa method was used to detect the levels of β-hexosaminidase and chymase in 1wocardial tissue. Results The expression levels of β-hexosaminidase, chymase and PKC in the Tongxinluo group were significantly lower than those in the model group ( P 〈 0. 01 ). Conclusion Tongxinluo can reduce the expression levels of β-hexosaminidase, chymase and PKC, inhibit mast cell de- granulation, alleviate inflammatory reaction of damaged myocardium, and then protect myocardial tissue.
