藤梨根制剂对卵巢癌细胞株A2780 MMP-2、MMP-9和TIMP-1表达的影响

摘　　要：目的探讨藤梨根制剂对卵巢癌细胞株A2780细胞基质金属蛋白酶2(MMP-2)、基质金属蛋白酶9(MMP-9)、组织金属蛋白酶抑制物1(TIMP-1)表达的影响,探讨藤梨根制剂抗卵巢癌机制。方法分别采用0 mg/ml、10 mg/ml、20 mg/ml、50 mg/ml、100 mg/ml藤梨根制剂处理卵巢癌细胞株A2780,培养24 h、48 h、72 h、96 h。采用噻唑蓝比色法(MTT)检测卵巢癌A2780细胞增殖情况,采用Transwell实验检测卵巢癌A2780细胞侵袭、迁移能力,采用实时荧光定量聚合酶链式反应(RT-PCR)及蛋白印迹法检测卵巢癌A2780细胞MMP-2、MMP-9、TIMP-1 mRNA及蛋白表达水平。结果 MTT法检测结果可见,10 mg/ml、20 mg/ml、50 mg/ml、100 mg/ml藤梨根制剂可明显抑制卵巢癌A2780细胞增殖,且呈现出一定的剂量依赖性、时间依赖性(P<0.05);20 mg/ml、50 mg/ml、100 mg/ml藤梨根制剂可明显降低卵巢癌A2780细胞迁移、侵袭能力,且呈现出一定的剂量依赖性(P<0.05);20 mg/ml、50 mg/ml、100 mg/ml藤梨根制剂可明显下调卵巢癌A2780细胞MMP-2、MMP-9表达,且呈现一定剂量依赖性(P<0.05),可上调卵巢癌A2780细胞TIMP-1表达。结论藤梨根制剂对卵巢癌A2780细胞增殖、迁移及侵袭能力具有明显抑制作用,推测其机制为下调卵巢癌A2708细胞MMP-2、MMP-9表达及上调TIMP-1表达。Objective To explore the influence of tengligen mixture on cell matrix metalloproteinase 2（MMP-2）,matrix metalloproteinase 9（MMP-9）,tissue inhibitor of metalloproteinase 1（TIMP-1） expressions in ovarian cancer cell line A2780,and explore the mechanism of tengligen mixture against ovarian cancer. Methods Ovarian cancer cell line A2780 was dealed by using tengligen mixture of 0 mg/ml,10 mg/ml,20 mg/ml,50 mg/ml,100 mg/ml. The proliferation of ovarian cancer A2780 cells was detected by using Thiazole blue colorimetric assay（MTT）. Invasion and migration ability of ovarian cancer A2780 cells was detected by using Transwell test. The expression levels of MMP-2,MMP-9,TIMP-1,mRNA and protein in ovarian cancer A2780 cells were tested by using real-time quantitative polymerase chain reaction（RT-PCR） and Western blotting. Results MTT assay results showed that tengligen mixture of 10 mg/ml,20 mg/ml,50 mg/ml,100 mg/ml can obviously inhibit the proliferation of ovarian cancer A2780 cells,and showed a dose dependent and time dependent manner（P〈0. 05）. Tengligen mixture of 20 mg/ml,50 mg/ml,100 mg/ml can significantly reduce migration and invasion ability of ovarian cancer A2780 cells,and showed a dose dependent manner（P〈0. 05）. Tengligen mixture of 20 mg/ml,50 mg/ml,100 mg/ml can down-regulate the expressions of MMP-2,MMP-9 in ovarian cancer cell line A2780,showing a dose dependent manner（P〈0. 05）,and it can up-regulate TIMP-1 expression in ovarian cancer cell line A2780. Conclusion It has obvious inhibitory effect of tengligen mixtureon on the proliferation,migration and invasion of ovarian cancer cell line A2780. It predicts that the mechanism is to down-regulate the expressions of MMP-2,MMP-9 in ovarian cancer cell line A2780 and up-regulate TIMP-1 expression.
