摘 要:目的研究黄连素对缺氧复氧诱导的心肌细胞凋亡的影响。方法以心肌细胞H9C2为研究对象,分为3组,依次为对照组、模型组、黄连素组,其中模型组和黄连素组进行缺氧复氧处理,黄连素组用黄连素预处理,对照组不做处理。MTT检测细胞增殖,流式细胞仪检测细胞凋亡,试剂盒检测乳酸脱氢酶(LDH)、丙二醛(MDA)、超氧化物歧化酶(SOD)含量,Western blot检测活化的含半胱氨酸的天冬氨酸蛋白水解酶3(Cleaved Caspase-3)、信号转导与转录因子3(STAT3)、磷酸化的STAT3(p-STAT3)表达。结果模型组细胞存活率、p-STAT3水平、SOD含量均明显低于对照组,而细胞凋亡率、LDH含量、MDA含量和Cleaved Caspase-3水平均明显高于对照组(P<0.05)。黄连素组细胞存活率、p-STAT3水平、SOD含量均明显高于模型组,而细胞凋亡率、LDH含量、MDA含量和Cleaved Caspase-3水平均明显低于模型组(P<0.05)。结论黄连素能够部分抑制缺氧复氧诱导的心肌细胞凋亡,并且对缺氧复氧抑制心肌细胞活性具有拮抗作用,其作用机制可能与细胞中STAT3信号通路有关。Objective To study the effect of berberine on apoptosis induced by hypoxia reoxygenation. Methods H9C2 was used as the research objects,which were divided into 3 groups,control group,model group,berberine group,Model group and berberine group were treated with hypoxia and reoxygenation,berberine group pretreatment with berberine,control group was not treated. Cell proliferation was detected by MTT,cell apoptosis was detected by flow cytometry,detection of lactate dehydrogenase( LDH),Malondialdehyde( MDA),Superoxide dismutase( SOD) content by kit,detection of Cleaved Caspase-3,STAT3 and p-STAT3 expression by Western blot. Results The cell survival rate,p-STAT3 level and SOD content in the model group were significantly lower than those in the control group,the apoptosis rate,LDH content,MDA content and Cleaved Caspase-3 levels were significantly higher than those in the control group( P〈0. 05). The cell survival rate,p-STAT3 level and SOD content in Berberine group were significantly higher than those in model group,the apoptosis rate,LDH content,MDA content and Cleaved Caspase-3 levels were significantly lower than the model group( P〈0. 05). Conclusion Berberine can partly inhibit the apoptosis of myocardial cells induced by hypoxia reoxygenation,and inhibit the activity of myocardial cells,which may be related to the STAT3 signaling pathway.
