摘  要:以彩叶凤梨继代苗为试材,通过甲基磺酸乙酯(EMS)诱变,研究了EMS处理对彩叶凤梨试管苗成活率及分化率的影响,同时以脯氨酸类似物L-羟脯氨酸(L-HYP)作为选择压力进行离体筛选,获得了彩叶凤梨抗羟脯氨酸的试管苗,并再生培养成抗性植株。结果表明,采用0.5%EMS对彩叶凤梨继代苗进行诱变处理6 h,其存活率和不定芽分化率分别为48.4%和36.9%;且7 mmol/L HYP筛选突变体植株效果最好,突变体植株在4℃低温胁迫下,超氧化物歧化酶(SOD)和过氧化物酶(POD)活性增强,叶片电导率降低,对低温环境表现出一定抗性。In this paper, the effect of EMS treatment on the survival rate and differentiation rate of tube seedlings in vitro from Neoregelia caroline was studied. And L-hydroxyproline(L-HYP)was used as selection stress to in vitro selection the mutants. Finally,hydroxyproline-resistant regenerated plants were obtained. The results showed that after 0.5% EMS treatment for 6 h, the survival rate and adventitious bud differentiation rate were 48.4% and 36.9%, respectively. The 7 mmol/L HYP screening mutants had the best effect,then the induced mutant could resist cold stress by increasing SOD and POD enzyme activities, decreasing conductivity under 4 ℃ low temperature stress.
