摘  要:目的:建立UPLC-MS/MS测定雪上一枝蒿甲素血液微透析探针回收率的分析方法。方法:采用浓度差法(增量法,减量法)测定血液探针体外回收率,并考察灌流液流速、浓度、温度对回收率的影响;反透析法(即减量法)测定其体内回收率,以探讨血液微透析技术用于雪上一枝蒿甲素体内药动学研究的可行性。结果:该试验建立的UPLCMS/MS分析方法在要求范围内线性良好,色谱的专属性、回收率、精密度等均符合微透析样品的研究要求。体外回收率试验,增量法和减量法测得回收率无显著差异;相同条件下,探针的体外回收率随着流速的增加而降低,随着温度的升高而明显增加,与探针周围雪上一枝蒿甲素的浓度无关;反透析法测得血液探针体内回收率是(52.9±3.76)%,探针日内回收率在12 h内稳定。结论:血液微透析探针可用于雪上一枝蒿甲素药动学研究,反透析法即减量法可用于雪上一枝蒿甲素体内回收率的测定。Objective:This study established UPLC-MS/MS method for determination of the blood microdialysis recovery of Bullatine A. Methods:By using the concentration difference method( incremental method,a reduction method),we measured in vitro recoveries and examined perfusate flow rate,concentration,temperature impact on the recovery of microdialysis and determined in vivo recovery by reverse dialysis method to explore the feasibility of blood microdialysis used for the pharmacokinetic study of Bullatine A. Results:In vitro recovery test,the method of UPLC-MS/MS was sensitive and reliable,the linearity correlation was good and the sample handling was quick,which was suitable for the determination of microdialysate sample. The recovery detected by increment method was the same as by decrement method. Under the same conditions in vitro recovery of the probe decreases with increasing flow rate,the recovery was independent of Bullatine A concentrations in the medium,but the Bullatine A recovery significantly increased as the temperature rose. The recovery rate of the blood probe was(52. 9 ± 3. 76) %,and the recovery of the probe was stable within 12 hours. Conclusion:The results showed that the blood microdialysis sampling can be used for the pharmacokinetic study of Bullatine A and retrodialysis can be used for the determination of the Bullatine A in vivo recovery.
