摘 要:目的:探讨无梗五加果实乙醇和多糖提取物对肝癌H22荷瘤小鼠的抗肿瘤活性。方法:采用将H22肝癌细胞接种于小鼠右前肢腋窝皮下的方法,建立小鼠肝癌H22肿瘤模型。随机设立空白组、模型组、阳性药对照组、无梗五加果实醇提物高、低剂量组、无梗五加果实粗多糖高、低剂量组、无梗五加果实精多糖高、低剂量组。ELISA法检测荷瘤小鼠血清中白细胞介素2(IL-2)、白细胞介素6(IL-6)、肿瘤坏死因子-α(TNF-α)含量。并称定瘤重、胸腺和脾脏,计算各给药组的抑瘤率、胸腺指数和脾脏指数。结果:与模型组相比,无梗五加醇提物高、低剂量组,粗多糖高剂量组,精多糖高、低剂量组对小鼠移植性实体瘤H22均有抑制作用,抑瘤率分别为53.27%、52.87%、51.98%、31.64%、38.14%,其中以无梗五加果实醇提物抑瘤作用最显著。且无梗五加果实各给药组(除粗多糖低剂量组)均能提高荷瘤小鼠的胸腺指数、脾脏指数及血清中IL-2、IL-6、TNF-α的水平,具统计学意义(P〈0.05)。结论:无梗五加果实乙醇提取物和多糖对小鼠H22移植性实体瘤都具较好的抑制作用,且乙醇提取物对肿瘤的抑制作用要强于多糖。Objective:To study the antitumor activity of the extracts of Acanthopanax sessiliflorus on H22 hepatoma bearing mice. Methods:H22 tumor-bearing mice model was induced by H22 cell vaccination in normal mouse forelimb right axillary subcutaneous. Normal group,model group,positive control group,the high and low dose groups of alcohol extract,the high and low dose groups of crude polysaccharide,the high and low dose groups of refined polysaccharide were randomly set up. The levels of IL-2,IL-6 and TNF-α in tumor-bearing mice were investigated by ELISA assay,the tumor weight and the tumor inhibition rate were calculated,the weight of thymus and spleen were weighted,the indexes of thymus and spleen were calculated. Results:Conclusion:Compared with model group,the high dose and low dose groups of ethanol extracts,the high dose groups of crude polysaccharides,the high dose and low dose groups of refined polysaccharides showed markably inhibitory effects on tumor growth,the inhibition rates were 53. 27%,52. 87%,51. 98%,31. 64% and 38. 14% respectivly. The ethanol extracts showed the strongest activity. Except the low dose group of crude polysacchride,other groups could increase spleen index and thymus gland index and enhance the expression levels of IL-2,IL-6,and TNF-α in tumor-bearing mice and the differences were statisticaly significant(P〈0. 05). Conclusions Both the ethanol extracts and polysaccharides of Acanthopanax sessiliflorus can inhibit tumor growth of H22 tumor-bearing mice. And the effect of ethanol extract was stronger.
