摘  要:本试验旨在研究程序性坏死阻断剂Necrostatin-1(Nec-1)对脂多糖(LPS)刺激断奶仔猪血细胞分类计数和血液生化指标的影响。选用25头断奶仔猪,分为对照组[10%二甲基亚砜(DMSO)+生理盐水]、LPS组(10%DMSO+LPS)、0.33Nec-1组[0.33 mg/kg体重(BW)Nec-1+LPS]、1.0Nec-1组(1.0 mg/kg BW Nec-1+LPS)和3.3Nec-1组(3.3 mg/kg BW Nec-1+LPS)。注射10%DMSO或Nec-1 30 min后,对照组注射生理盐水,其他组注射100μg/kg BW LPS,分别在注射LPS或生理盐水4 h和24 h后采血液样品待测。结果表明:注射LPS 4 h后,0.33 mg/kg BW Nec-1、1.0 mg/kg BW Nec-1和3.3 mg/kg BW Nec-1显著缓解LPS刺激导致的血小板分布宽度升高(P<0.05);0.33 mg/kg BW Nec-1能显著缓解LPS刺激导致的血浆谷草转氨酶(AST)活性和AST/谷丙转氨酶(ALT)比例的升高(P<0.05),1.0 mg/kg BW Nec-1显著缓解血浆葡萄糖含量和肌酸激酶活性的降低(P<0.05);注射LPS 24 h后,0.33 mg/kg BW Nec-1、1.0 mg/kg BW Nec-1和3.3 mg/kg BW Nec-1可显著缓解LPS刺激导致的嗜酸性粒细胞比例的升高(P<0.05),并有缓解嗜酸性粒细胞绝对值升高的趋势(P<0.10);0.33 mg/kg BW Nec-1缓解了肌酸激酶活性的降低并提高了血浆总胆固醇和甘油三酯含量(P<0.05),1.0 mg/kg BW Nec-1有缓解LPS刺激导致的血浆AST/ALT升高的趋势(P<0.10),3.3 mg/kg BW Nec-1能显著缓解AST/ALT比例的升高(P<0.05)和葡萄糖的降低(P<0.05),并有缓解AST升高和总胆固醇降低的趋势(P<0.10)。以上结果表明,Nec-1对LPS刺激导致的免疫应激反应具有一定的缓解作用。The study was conducted to investigate the effects of Necrostatin-1(Nec-1) on blood cell differential count and blood biochemical measurements of piglets challenged with lipopolysaccharide(LPS). Twenty-five piglets were randomly assigned into 5 groups: control; LPS; LPS+0.33 Nec-1(0.33 mg/kg BW); LPS+1.0 Nec-1(1.0 mg/kg BW); LPS+3.3 Nec-1(3.3 mg/kg BW). After injection of 10%DMSO or Nec-1 for 30 mins, piglets were injected with either LPS or saline. At 4 h or 24 h after LPS or saline injection, blood samples were taken. The results showed that at 4 h, 0.33 mg/kg BW Nec-1, 1.0 mg/kg BW Nec-1 and 3.3 mg/kg BW Nec-1 alleviated the decrease of platelet distribution width induced by LPS challenge(P0.05). 0.33 Nec-1 alleviated the increases of aspartate transaminase(AST) and AST/alanine aminotransferase(ALT)(P0.05), and 1.0 Nec-1 alleviated the decrease of blood glucose and creatine kinase(P0.05) induced by LPS challenge. At 24 h, Nec-1 alleviated the increase of eosinophil proportion( mg/kg BW P0.05), and had a tendency to alleviate the increase of eosinophil count(P0.10) induced by LPS challenge. 0.33 mg/kg BW Nec-1 alleviated the decrease of creatine kinase and increased total cholesterol and triglyceride levels(P0.05). 1.0 Nec-1 had a tendency to alleviate the increase of AST/ALT induced by LPS challenge(P0.10). 3.3 mg/kg BW Nec-1 alleviated the decrease of blood glucose and the increase of AST/ALT(P0.05), and had a tendency to alleviate the decrease of AST and total cholesterol(P0.10) induced by LPS challenge. These results indicate that Nec-1 alleviated the stress response induced by LPS challenge.
