摘  要:【目的】以菜心抗小菜蛾品种Caixin65和感小菜蛾品种Caxixin69及其杂交后代所得的54份F2∶3家系为实验材料,利用SCo T标记检测亲本及子代分离群体的遗传多样性,为新品种选育和分子标记辅助育种提供基础材料和理论基础。【方法】利用筛选的17条SCo T引物检测54份菜心F2∶3家系及2个亲本的遗传多样性。【结果】17对引物共扩增出条带161条,其中多态性条带共61条,多态性条带百分率为38.80%;平均每条引物扩增出条带9条,其中多态性条带4条。平均多态性位点百分率为45.02%,平均观察等位基因数(Na)为1.1613;平均有效等位基因数为1.3184;平均Shannon多样性指数(I)为0.2723;平均期望杂合度(He)为0.1865;平均Nei遗传距离为0.225。表明多数家系的分子遗传距离相近,F2∶3家系的遗传多样性较低,但少数家系之间已经发生了遗传多样性变化。在Nie遗传距离0.080之处,可以将56份菜心分为5类。SCo T标记没有检测出多数家系间的显著的基因组变化,少数家系呈现出Sco T多态性,表明SCo T标记可以继续用于优异家系选育。【结论】SCo T标记具有操作简单、重复性高、多态性较丰富等优点,通过SCo T检测表明大部分菜心F2∶3家系间遗传距离接近,但少数家系间已经发生了分离,将SCo T标记用于菜心F2分离群体的遗传多样性检测,结合表型筛选将有助于品种选育效率的提高。【Objective】In order to provide basic material and theory for breeding of new variety and molecular markers-assisted breeding,SCo T markers were used to assess the genetic diversity of Chinese flowering cabbages varieties which included Caixin65,Caixin69 and 54 F2∶ 3 family lines obtained from the hybrid offspring of these two varieties.【Method】The genetic diversity of 54 F2∶ 3 family lines and two parents were assessed by 17 selected SCo T primers. 【Result】A total of 161 DNA bands were amplified by 17 SCo T primers of which 61 were polymorphic bands. The mean percentage of polymorphic bands was 38. 80 %. The mean polymorphic bands were 9 for each primer of which4 were polymorphic bands. The fragment sizes ranged from 500 bp to 1500 bp. The mean percentage of polymorphic loci was 45. 02 %,the mean number of different alleles( Na) was 1. 1613,the number of effective alleles( Ne) was 1. 3184,Shannon's Information Index( I)was 0. 2723 and Nei's gene diversity( He) was 0. 1865. The mean Nei's genetic distance was 0. 225. The 56 Chinese flowering cabbages were grouped 5 clusters at the Nei's genetic distance 0. 080 based on UPGMA approach. The genetic distances of most lines were closer,it meant that there was not significant genomic variation between most family lines. While SCo T polymorphism showed rich in minority family lines indicated that it could be continually used in excellent lines selection. 【Conclusion】The SCo T marker has many advantages which is simple to operate and high repeatability and rich polymorphism,etc. The genetic diversity in most of 54 F2∶ 3 family lines are closely but less being separate.So SCo T marker will improve the efficiency of breeding through genetic diversity accession and phenotypic screen.
