摘 要:为了建立快速提取检测雷公藤甲素的分析方法,本试验研究了雷公藤根皮、不定根和发状根中雷公藤甲素的超临界CO_2(SFE-CO_2)萃取工艺,并使用液相色谱质谱联用仪(HPLC-MS)检测雷公藤甲素含量。结果表明:超临界CO_2萃取根皮中甲素较适宜条件为:萃取压力48.265 MPa,萃取温度35℃,改性剂正丁醇、静态萃取时间15min及CO_2体积25 mL。超临界CO_2萃取不定根和发状根中甲素较适宜条件为:萃取压力27.580 MPa,萃取温度55℃,改性剂乙醇、静态萃取时间20 min及CO_2体积25 mL。添加回收率为80%~107%,相对标准偏差(RSD)为7.7%~10.6%。研究表明,超临界CO_2萃取全程分析时间小于1h,与常规溶剂法相比,该方法具有萃取效率高、无污染、经济等优点。In order to establish an analytical method for rapid extraction and detection of triptolide,the supercritical CO_2(SFE-CO_2)extraction process of triptolide in roots,adventitious roots and hairy roots of Tripterygium wilfordii were studied.The instrument(HPLC-MS)was used to detect the content of triptolide.The results showed that the optimum conditions for the extraction of triptolide in the root bark of supercritical CO_2 were as follows:extraction pressure was 48.265 MPa,extraction temperature was 35℃,modifier was n-butanol,static extraction time was 15 min and CO_2 volume was 25 mL.Supercritical CO_2 extraction of adventitious roots and hairy roots of triptolide suitable conditions were:extraction pressure was27.580 MPa,extraction temperature was 55 ℃,modifier was ethanol,static extraction time was 20 min and CO_2 volume was 25 mL.The recovery was 80% to 107%,and the relative standard deviation(RSD)was 7.7%to 10.6%.The research showed that the whole analysis time of supercritical CO_2 extraction was less than 1 h.Compared with the conventional solvent method,the method has the advantages of high extraction efficiency,no pollution and economy.
