摘 要:目的探讨高原环境中妊娠期不同程度缺氧对胎羊心肌组织中基质金属蛋白酶8(MMP-8)表达的影响。方法选取24只平原环境的妊娠期绵羊,按照简单随机抽样法分为平原对照组(8只)、轻度低氧组(8只)和重度低氧组(8只),应用人工实验舱模拟高海拔环境,制作不同程度缺氧的妊娠期动物模型,采用扫描电子显微镜观察胎羊心肌组织和线粒体的病理超微结构的改变,Masson染色评价胎羊心肌纤维化程度,蛋白免疫印迹法观察MMP-8蛋白表达变化。结果扫描电子显微镜显示,轻度低氧组和重度低氧组心肌组织排列出现不同程度紊乱,线粒体形态结构和分布出现不同程度的异常和混乱,其中重度低氧组可见部分肌纤维断裂重新分布,出现"巨线粒体";Masson染色显示,轻度低氧组(2. 165%±0. 028%)和重度低氧组(4. 612%±0. 035%)纤原面积百分比明显增大,且重度低氧组高于轻度低氧组,差异有统计学意义(P <0. 05);Western blot结果显示,轻度低氧组(5. 87%±1. 32%)和重度低氧组(4. 83%±0. 59%)的MMP-8蛋白相对表达量明显降低,且重度低氧组低于轻度低氧组,差异有统计学意义(P <0. 05)。结论高原海拔升高缺氧加剧可能通过下调MMP-8蛋白表达对胎羊心肌组织及线粒体造成损伤。Objective To investigate the influence on expression of matrix metalloproteinase-8 (MMP-8) protein of sheep embryonal myocardium during pregnancy at different levels of hypoxia in high altitude condition. Methods A total of 24 maternal sheep in low altitude condition were divided into three groups randomly:low altitude control group(LAC, n =8),mild hypoxia exposure group(MHE, n =8)and severe hypoxia exposure group(SHE, n =8).High altitude condition was simulated by artificial experimental cabin to make pregnant animal models with different level of hypoxia.The myocardial ultra-structural changes and mitochondria of sheep embryo were observed by scanning electron microscopy(SEM).Degree of myocardial fibrosis was evaluated by Masson staining and Image pro plus analysis system and expression of MMP-8 protein was observed by Western blotting. Results SEM indicated that derangement of myocardial tissue,mitochondrial morphology change and distribution in MHE and SHE group.Redistribution subsequent to rupture of myocardial fiber and the mega-mitochondria was seen in SHE group.Masson staining showed that the percentage in area of myocardial collagen in MHE and SHE group increased,which was more obvious in SHE group compared with MHE group with significant difference(4.612%±0.035% vs. 2.165%±0.028%, P 〈0.05).Western blotting showed that the expression of MMP-8 protein decreased in MHE and SHE group,which was lower in SHE group compared with MHE group with significant difference(4.83%±0.59% vs. 5.87%±1.32%, P 〈0.05). Conclusions High altitude induced hypoxia may decrease expression of MMP-8,which may cause mitochondrial damage of sheep embryonal myocardium during pregnancy.
