摘  要:目的观察不同浓度虫草素对人宫颈癌Hela细胞增殖与凋亡的影响。方法不同浓度虫草素处理Hela细胞24 h,倒置显微镜观察细胞形态变化,CCK-8法检测细胞增殖活性,流式细胞术检测细胞凋亡,Western-blot检测凋亡相关蛋白。结果 Hela细胞增殖活性随虫草素浓度增加而降低(P<0.05),虫草素引起Hela细胞早期凋亡(P<0.05),降低Bcl-2蛋白、增加Bax蛋白表达,诱导Cleaved-PARP裂解。结论虫草素通过下调Hela细胞中Bcl-2并上调Bax表达,促进早期凋亡。Objective To observe the effect of different concentrations of cordycepin on the proliferation and apoptosis of human cervical carcinoma Hela cells. Methods Hela cells were treated with different concentrations of cordycepin for 24 h, and their morphology, proliferation, apoptosis, and apoptosis-related protein levels were detected inverted microscopy, CCK-8, flow cytometry, and Western blot, respectively. Results The proliferative activity of Hela cells was decreased as incremental concentrations of cordycepin(P〈0.05). Cordycepin induced the early apoptosis(P〈0.05) and Bax and cleavedPARP expression, but reduced Bcl-2 expression in Hela cells. Conclusion Cordycepin can promote early apoptosis via downregulating Bcl-2 and upregulating Bax expression in Hela cells.
