摘  要:目的明确三叶青水、乙醇、乙酸乙酯三种提取物体外对乳腺癌细胞(MCF-7)的抑制作用,分析不同提取方式对肿瘤细胞抑制强弱差异。方法 MTT法检测三叶青三种提取物含药血清分别在2、10、50、100、200μg·m L-1时对MCF-7细胞存活率的影响;采用流式细胞仪检测三种提取物含药血清在200μg·m L-1浓度时对MCF-7细胞凋亡的影响。结果三种提取物均具有与浓度相关的MCF-7细胞体外抑制作用(P<0.05),作用强弱依次是乙酸乙酯提取物>乙醇提取物>水提取物;三种提取物均能促进MCF-7细胞凋亡(水、乙醇组P<0.05,乙酸乙酯组P<0.01);乙酸乙酯提取物比水提物组、乙醇组具有更强的凋亡促进作用(P<0.05)。结论三种提取物均具有体外抗乳腺癌MCF-7细胞作用,其中乙酸乙酯提取物作用最强。Objective To clarify the inhibitolT effect of three extracts from radix tetrastigme water, ethanol and ethyl acetate on breast cancer cells (MCF-7) , and to analyze the difference of different extraction sites on tumor cells. Methods MTT method of three kinds of green leaf extract serum respectively in 2, 10, 50, 100, 200μg.mL^-1 on the survival rate of MCF-7 cells detected by three kinds of effects was made. The effect of extract serum at 200μg.mL^-1 concentration of MCF-7 cell apoptosis by flow cytometry was carried out. Results The three extracts had MCF-7 cells in vitro and concentration dependent inhibition (P〈0.05) , the intensity of the effects followed by ethyl acetate extract ethanol extract, water extract; three extracts could promote the apoptosis of MCF-7 cells (water, ethanol and ethyl acetate group P〈0.05, acetic ether group P〈0.01) , ethyl acetate extract than water extract group and ethanol group has stronger apoptosis (P〈0.05) . Conclusion all of the three kinds of extracts have the effect of anti breast cancer MCF-7 cells in vitro, and the ethyl acetate extract has the strongest effect.
