摘  要:目的研究转化生长因子-β1(TGF-β1)和IL-6在野百合碱诱导的肺动脉高压(PAH)大鼠肺组织中的表达水平及意义。方法 22只雄性SD大鼠随机分为正常对照组(n=6)、野百合碱-3周组(MCT-3w组,n=8)和MCT-5w组(n=8),皮下注射野百合碱复制肺动脉高压模型。检测右心室收缩压(RVSP)和右心室肥厚指数(RVHI),左肺组织经固定包埋切片后行van Geison染色,光镜下观察肺内小动脉病理改变,计算肺内小动脉血管壁厚度占血管直径的百分比(WT%)和血管壁面积占小动脉总横截面积的百分比(WA%)。蛋白免疫印迹法检测右肺组织TGF-β1表达水平,ELISA检测肺组织中IL-6表达水平。结果与正常对照组相比,MCT-3w组和MCT-5w组大鼠的RVSP和RVHI均升高(P<0.05),肺小动脉中膜增厚,WT%和WA%均增加(P均<0.05),肺组织中TGF-β1和IL-6表达水平均上调(P均<0.05);与MCT-3w组相比,上述指标在MCT-5w组增高更明显(P均<0.05)。结论 TGF-β1和IL-6在野百合碱诱导的肺动脉高压大鼠肺组织中的表达水平均增高,TGF-β1和IL-6可能参与了PAH的发病和进展。Objective To investigate the expression level and significance of transforming growth factor-beta 1(TGF-β1) and interleukin-6 (IL-6)in the lung tissues of rat models with pulmonary hypertension induced by monocrotaline. Methods Twenty-two male SD rats were randomly divided into the normal control group(n=6), 3-week monocrotaline group(MCT-3w group, n=8)and 5-week monocrotaline group(MCT-5w group, n=8). Pulmonary hypertension rat models were established by subcutaneous injection of monocrotaline. The right ventricular systolic pressure(RVSP)and right ventricular hypertrophy index(RVHI)were measured. The left lung tissue sections were stained with van Geison staining. The pathological changes of the pulmonary arterioles were observed under optical microscope. The percentage of wall thickness (WT%) of pulmonary arterioles to the vascular diameter and wall area (WA%) to the total cross-sectional area of pulmonary arterioles was calculated. The expression level of TGF-β1 in the right lung tissues was quantitatively detected by Western blot. The concentration of IL-6 in the lung tissues was measured by ELISA. Results Compared with the normal control group, the RVSP and RVHI were significantly elevated (both P 〈 0.05), the middle membrane of pulmonary arterioles was considerably thickened, the WT% and WA% were significantly increased (both P 〈 0.05) and the expression levels of TGF-β1 and IL-6 in the lung tissues were evidently up-regulated (both P 〈 0.05) in the MCT-3w group and MCT-5w group. Compared with the, the parameters above were significantly increased in the MCT-5w group (all P 〈 0.05). Conclusions The expression levels of TGF-β1 and IL-6 are significantly up-regulated in the lung tissues of rats with pulmonary hypertension induced by monocrotaline. TGF-β1 and IL-6 may be involved in the pathogenesis and progression of pulmonary hypertension.
