摘  要:目的:研究附子对NOS抑制小鼠及其亚硝酸钠-NO通路的影响。方法:采用持续10天灌胃给予L-NNA 700mg/kg·d^(-1)的方法制备高血压小鼠模型,将小鼠随机分为7组,每组20只,分别给予1%CMC-Na溶液、L-NNA、L-NNA+附子水煎剂(4g/kg)、L-NNA+附子水煎剂(4g/kg)+别嘌呤醇(70mg/kg·d^(-1))、L-NNA+亚硝酸钠(0.25mg/kg·d^(-1))、L-NNA+亚硝酸钠(0.25mg/kg·d^(-1))+别嘌呤醇(70mg/kg·d^(-1))、L-NNA+别嘌呤醇(70mg/kg·d^(-1))。测定各组小鼠给药10天后血压,心、肾组织重量,检测心、肾组织和血清中NO和MDA含量。结果:亚硝酸钠和附子均可减弱由NOS抑制剂诱导的高血压效应,别嘌呤醇虽无法影响模型小鼠的血压,但可以削弱附子、亚硝酸盐的降压作用。黄嘌呤氧化还原酶在亚硝酸盐向NO转化过程中起着重要的催化作用,同时黄嘌呤氧化还原酶在附子升高小鼠体内NO含量中起重要作用。附子和亚硝酸钠均可降低NOS抑制小鼠体内氧化应激的程度,黄嘌呤氧化还原酶抑制剂并不干扰其作用。结论:附子对NOS抑制小鼠的降压作用可能是通过亚硝酸盐-NO通路实现的。Objective:To study the Effects of Radix Aconiti lateralis preparata on nitrite-no pathway of NOS Inhibition Mice.Methods:The Nitric oxide synthase(NOS)inhibition-induced hypertension model was established by intragastric administration with L-NNA(700 mg/kg)for ten days.ICR mice pretreated 1%CMC-Na,nitric oxide synthase(NOS)inhibitor L-NNA,L-NNA +XOR inhibitor allopurinol,L-NNA + Radix Aconiti lateralis preparata,L-NNA + nitrite,L-NNA + Radix Aconiti lateralis preparata+ allopurinol,L-NNA + nitrite+ allopurinol(15 mice per group)for ten days.The contents of NO and the contents of MDA in heart,kidney and plasma were assayed after 10 days.Study the pathological changes of kidney and heart in mice,and survey the changes of blood pressure.Results:nitrite and Radix Aconiti Lateralis Preparata can attenuate the hypertensive effect induced by NOS inhibitor.The vasodilator responses to Radix Aconiti lateralis preparata were attenuated by allopurinol.Xanthine oxidation-reductase plays an important catalytic role in the conversion process of nitrite to NO.Meanwhile,Xanthine oxidation-reductase plays an important role in the elevation of NO content induced by Radix Aconiti lateralis preparata.Radix Aconiti Lateralis Preparata and nitrate significantly decreased the content of MDA in heart,kidney compared with the model group.Xanthine oxidoreductase had nothing to do with the antioxidant effect of Aconiti lateralis preparata and nitrate.Conclusion:The hypotensive effect of Radix Aconiti Lateralis Preparata on NOS Inhibition Mice was conducted through the nitrite-NO pathway.
