摘  要:目的:优化水杨梅总黄酮半仿生酶法提取工艺。方法:以总黄酮含量为指标,比较乙醇回流法、半仿生法、半仿生酶法提取水杨梅中总黄酮的效果。用响应面法优化半仿生酶法提取方法,以料液比、提取温度、提取时间为因素,在单因素试验的基础上,采用三因素三水平的响应面试验,优化工艺参数。结果:在三种提取方法中,以半仿生酶法得到的总黄酮含量最高,优化水杨梅半仿生酶法最佳提取工艺条件为:料液比1∶10、提取温度50℃、提取时间1h,在此条件下水杨梅中总黄酮平均提取量10.1770mg。三因素对水杨梅总黄酮提取率影响主次顺序为提取温度>提取时间>料液比。结论:优化后的半仿生酶法提取水杨梅中总黄酮,提取率高且工艺稳定。Objective:To optimize the extraction process of total flavonoids with semi-bionic enzyme method.Methods:Taking total flavonoids content as the index,compare the extraction effect of the total flavonodis from the Andia rubella Hance by ethanol reflux method,semi-bionic method and semi-bionic enzyme method.With liquid ratio,extraction temperature,extraction time as factor,based on single factor experiments,response surface experiments were used to optimize the extraction method.Results:In the three extraction methods,semi-bionic enzyme had highest total flavonoids content,the best extraction condition was 1:10 of the ratio of material to solution,extraction temperature 50℃.extraction time 1 h,the conditions for launching arbutus flavonoids extraction yield an average of 10.1770 mg.The effect of three factors on the extaction rate of the total flavonoids was extraction temperature,extraction time,the ratio of material to solution.Conclusion:The optimized semi-bionic enzyme method showed high extraction rate and stable process.
