摘 要:目的以模式生物斑马鱼为动物模型研究乌头碱的神经毒性并初步探索其神经毒性的致病机制。方法应用不同浓度的乌头碱(0μM,0.1μM,0.5μM,1μM,10μM,100μM)作用胚胎期0-4天后,观察5-8天斑马鱼幼鱼游泳行为的改变;在相差显微镜下观察幼鱼肌节发育和胚胎早期感觉神经元Rohon-Beard(RB)神经元发育情况,并测定不同浓度乌头碱对胚胎脊索乙酰胆碱酯酶活性的作用。结果乌头碱以剂量依赖的方式降低斑马鱼幼鱼的游泳活力,与对照组相比较,乌头碱浓度大于0.5μM即可显著性降低斑马鱼游泳活力;可观察到幼鱼的肌节长度与对照组比较也有显著性降低;并发现随着乌头碱浓度的增加,BR神经元的生理性凋亡受到抑制。检测不同浓度乌头碱作用于斑马鱼幼鱼24h、48h、72h后的脊索匀浆乙酰胆碱酯酶的活性,发现乌头碱作用72h后,随着乌头碱浓度的增加,乙酰胆碱酯酶活性显著降低。结论乌头碱通过抑制乙酰胆碱酯酶活性,对斑马鱼胚胎和幼鱼发育产生神经毒性。Objective To investigate the influence of aconitine solution on the embryonic development of zebrafish and neurobehavior of juvenile fish.Methods The embryos of zebrafish at 6 hours after fertilization were exposed to aconitine solution at a concentration of 0μM,0.1μM,0.5μM,1μM,10μM,100μM.Quantitative analysis of behavior,histometrycal and cellular morphology,AChE activity were performed.Results Results showed that aconitine could weakened the swimming activity of juvenile fish in dose response manner.The behavioral changes caused by aconitineinduced hypogenetic axial muscles Another interesting phenomenon was that embryonic Rohon-Beard(RB)cells survived the physiological apoptosis.Corresponded to above changes,AChE activity level has been down-regulated in aconitine treated juvenile fish Conclusion Aconitine exposure may cause embryonic developmental disorder in zebrafish and abnormal neurobehavior in juvenile fish.
