膈下逐瘀汤加减方对气滞血瘀型输卵管炎大鼠模型TNF-α蛋白表达的影响

摘　　要：目的研究应用多种刺激联合输卵管注菌的方法、制造出符合中医证候特点的气滞血瘀型输卵管炎大鼠模型,观察通过膈下逐瘀加减方治疗对该模型大鼠TNF-α蛋白表达的影响。方法将48只Wistar雌性大鼠随机分为4组,每组12只,分别为正常组、模型组(气滞血瘀型输卵管炎组)、成药组(康妇炎胶囊组)、中药组(膈下逐瘀汤加减方组)。正常对照组予以普通喂养,模型组给予输卵管注菌造模,并采用多种刺激干预处理以造成气滞血瘀型输卵管炎不孕大鼠模型,并观察大鼠输卵管及外部体征变化情况。成药组与中药组:即经过康妇炎胶囊以及膈下逐瘀汤加减方治疗后,观察气滞血瘀型输卵管炎大鼠的TNF-α蛋白表达情况的不同。结果模型组出现气滞血瘀型输卵管炎的症状,输卵管与周围组织黏连,显微镜下观察输卵管形态改变、增粗、积水,炎症指标TNF-α差异比较,有统计学意义(P<0.05)。结论多种刺激联合输卵管注菌建立的气滞血瘀型输卵管炎模型,符合输卵管炎病理特点和中医气滞血瘀证候的主要辨证要点,是较为理想的气滞血瘀型输卵管炎模型。膈下逐瘀汤加减方能抑制输卵管上皮TNF-α蛋白的表达,强度优于成药组。Objective To explore the method of combined tubal injection stimulation on the application of a variety of bacteria, producing model of arthritis in rats with qi stagnation and blood stasis syndrome characteristics of fallopian tube, and through the observation of Gexia Zhuyu decoction on the treatment effect of the expression of TNF-α protein in model rats. Methods 48 Wistar female rats were randomly divided into 4 groups, 12 in each group. They were normal group, model group（qi stagnation and blood stasis type salpingitis group）, medicine group（Kangfuyan capsule group）, Chinese medicine group（Gexia Zhuyu decoction）. The normal control group was given general feeding, the model group was given the salpingo injection, and the multiple stimulation intervention was applied to induce the tubal inflammation infertility model of qi stagnation and blood stasis type, and the changes of the oviduct and external signs were observed. After the treatment of Kangfuyan capsule and Gexia Zhuyu decoction, the expression of TNF-α protein in rats with salpingitis induced by qi stagnation and blood stasis was observed. Results In the model group, there were symptoms of salpingitis with qi stagnation and blood stasis, and the adhesion between the oviduct and the surrounding tissues. The morphological changes, thickening and hydropsis of the fallopian tubes and inflammatory index TNF-α were observed by microscope, and the difference was statistically significant（P〈0.05）. Conclusion The model of tubal inflammation with qi stagnation and blood stasis established by multiple stimulation combined with oviduct injection is consistent with the pathological characteristics of salpingitis and the main dialectical points of TCM qi stagnation and blood stasis syndrome. It is an ideal tubal inflammation model with qi stagnation and blood stasis. The expression intensity of TNF-α protein in tubal epithelium is better than that of the group of Kangfuyan capsule.
