针刺对血管性痴呆大鼠TNF-α水平及神经行为学的影响

摘　　要：目的研究针刺对血管性痴呆大鼠神经行为学和血清中肿瘤坏死因子-α(TNF-α)含量的影响,为针灸防治血管性痴呆提供实验依据。方法将90只Wistar雄性大鼠,随机分为3组,对照组、模型组和治疗组,采用双侧颈总动脉(CCA)结扎反复缺血再灌注(2VO)的方法制作血管性痴呆的模型,模型制作成功后以神经行为学评分的方法评价模型是否成功,然后治疗组予以针刺百会、四神聪、大椎治疗。于结束治疗3 d后同时对3组进行神经行为学评分,且不同时间点运用免疫组化法检测3组大鼠血清中TNF-α的含量。结果同一时间点治疗组大鼠血清中TNF-α含量明显低于模型组,并且神经行为学评分低于模型组,运用统计学软件进行统计学分析,其差异具有统计学意义(P<0.05)。结论针刺能明显降低血管性痴呆大鼠血清中的TNF-α含量,改善神经功能,具有显著的脑保护作用。Objective To study the effect of acupuncture on neurobehavior and content of TNF-α in serum of rats with vascular dementia, to provide experimental basis for acupuncture and moxibustion in the prevention and treatment of vascular dementia. Methods Ninety male Wistar rats were randomly divided into three groups： control group, model group and treatment group. Model of vascular dementia was established by bilateral common carotid artery（CCA） ligation and repeated ischemia-reperfusion（2 VO） method. After the model was evaluated by the method of neurobehavioral score and then the treatment group was treated with acupuncture in Baihui, Sishengcong and Dazhui. The neurobehavioral scores of the three groups were evaluated at the same time three days after the treatment and the content of TNF-α in the serum of the three groups of rats were detected by immunohistochemistry at different time points. Results The content of TNF-α in serum of treatment group was significantly lower than that of the model group at the same time point, and the neuroethology score was lower than that of the model group. Statistical analysis was performed using statistical software. The difference was statistically significant（P〈0.05）. Conclusion Acupuncture can obviously reduce the content of TNF-α in serum of vascular dementia rats and improve neurological function and has a significant protective effect on brain.
