摘 要:目的探讨Toll样受体(Toll-like receptor,TLR)2(T597C,rs3804099)基因多态性与军团菌宿主易感性的关系。方法分离健康人外周血单个核细胞,用军团菌刺激24h后,采用酶联免疫吸附实验(enzyme-linked immunosorbent assay,ELISA)检测培养上清液中肿瘤坏死因子-α(tumor necrosis factor-alpha,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-6(interleukin-6,IL-6)的水平,real-time PCR检测TLR2及髓样分化因子88(myeloid differentiation factor 88,MyD88)基因水平,对TLR2(597T→C)多态性采用SNaPshot技术检测其不同基因型。结果 TLR2(C597T)CT/TT基因型个体PBMC中MyD88的表达水平低于TLR2(C597T)CC基因型(1.37,1.27;P=0.048);同时TLR2(C597T)CT/TT基因型个体TNF-α、IL-1β和IL-6水平高于CC基因型(880.08,740.67;P=0.046:948.09,657.01;P=0.042:4818.9,2083.8;P=0.012)。结论提示TLR2基因多态性与军团菌感染的宿主易感性有关,rs3804099CT/TT可能为军团菌病的保护性基因。OBJECTIVE To explore the relationship of Toll-like receptor 2(T597 C,rs3804099)gene polymorphism and the susceptibility of Legionella infection.METHODS PBMCs were isolated from healthy subjects,and were stimulated with L.pneumophila for 24 h.The levels of TNF-α,IL^-1βand IL-6 in culture supernatant were measured with enzyme-linked immunosorbent assay(ELISA)kit.The mRNA expression of TLR2 and MyD88 were detected by real-time PCR.TLR2 genotyping was performed with SNaPshot technology.RESULTS The MyD88 mRNA expression level in PBMCs was significantly lower with genotype TLR2(C597 T)CT/TT in comparison to the CC homozygotes(1.37,1.27;P=0.048).The levels of TNF-α,IL^-1βand IL-6 with genotypes TLR2 CT/TT homozygotes/heterozygotes were higher than those of the CC homozygotes(880.08,740.67;P=0.046:948.09,657.01;P=0.042:4818.9,2083.8;P=0.012).CONCLUSION These results suggest that TLR2 polymorphic individuals may be associated with susceptibility of L.pneumophilainfection.rs3804099 CT/TT may be a protective gene for L.pneumophila.
