摘 要:以川西南11个泡核桃优良品种资源为材料,利用SSR荧光标记技术进行了DNA指纹图谱构建和遗传关系分析。11对SSR荧光引物在11个泡核桃品种中共检测到59个等位基因,每个SSR位点有3~8个等位基因,平均为5.364个,有较好的多态性。期望杂合度(He)、香农信息指数(I)和多态信息含量(PIC)、变幅分别为0.579~0.851、0.993~1.972和0.490~0.833;均值分别为0.692、1.379和0.642。筛选出4对SSR引物wga004、wga032、wga070、zmz05为核心引物构建了各品种的指纹图谱。聚类分析中,11个泡核桃品种间遗传相似系数在0.525~0.932之间。说明11个泡核桃品种间遗传多样性较丰富,亲缘关系与选育的地理来源存在一定的相关性,地域上基因交流广泛,遗传背景复杂。Eleven improved cultivars of Juglans sigllata from southwestern Sichuan were used as materials to construct DNA fingerprinting and analyze genetic relationship with simple sequence repeats(SSR) fluorescent markers.A total of 59 alleles were detected with considerable polymorphism from 11 pairs of SSR fluorescent primers in 11 Juglans sigllata varieties,and the number of the alleles per SSR locus ranged from 3 to 8 with an average of5.364.The ranges of expected heterozygosities(He),shannon index(I) and polymorphism information content(PIC)were 0.579~0.851,0.993~1.972,and 0.490~0.833,respectively,with an average of 0.692,1.379 and 0.642 respectively.Four pairs of SSR primers(wga004,wga032,wga070 and zmz35) were selected as core primers and used for the DNA fingerprint construction of each variety.The genetic similarity coefficients of 11 Juglans sigllata cultivars were ranged from 0.525 to 0.932 in clustering analysis.The results indicated that genetic diversity is abundant among the 11 Juglans sigllata cultivars,and genetic relationship was generally correlated to their geographic origins of breeding.Regional gene exchange was extensive among the cultivars,and the genetic background was complex.
