摘 要:从分子水平研究铁皮石斛的遗传多样性,探讨不同铁皮石斛品种的亲缘关系的远近。为铁皮石斛品种的鉴定,后代的区分,新品种的鉴定和种质资源的利用与保护提供科学依据。本研究利用SRAP分子标记法对26份取自光明食品集团云南石斛生物科技开发有限公司资源圃的铁皮石斛品种的基因组DNA进行分析。从81对引物中共筛选出15对引物。该15对引物对26份材料共扩增出231条清晰的条带,其中185条具有多态性,占总条带数的80.09%。UPGMA聚类分析结果表明,在相似系数0.78处26份材料被划分为3个类群。研究结果表明SRAP分子检测技术能较好地用于铁皮石斛品种亲缘关系的研究,可用于铁皮石斛后代的区分,品种的鉴定。The genet ic diversity of Den drobium candidum was studied at molecular level,and the relationship between different Dendrobium officinale varieties was discussed,which could provide scientific reference for variety identification,offspring classification,new variety identification and protection and utilization of germplasm resources of Dendrobium officinale.The sequence-related amplified polymorphism(SRAP) was assessed to analyze the genomic DNA among 26 species of Dendrobium officinale from Guangming Yunnan Shihu Co.Ltd..15 pairs of primers were screened from 18 pairs of primers.A total of 231 DNA bands were amplified by 15 selective primers from 26 materials,185 of which were polymorphic,accounting for 80.09%.UPGMA clustering analysis indicated that 26 materials were classified into three cluster groups with the genetic similarity of 0.78.The results demonstrated that the SRAP molecular detection technology could be used to study the genetic relationship of Dendrobium candidum,which could be used to distinguish the offspring and identify the varieties of Dendrobium candidum.
