摘 要:香果树是中国特有珍稀濒危树种,为建立一种高频香果树再生体系,以香果树叶片和叶柄为外植体,利用含新型植物激素TDZ和NAA的培养基诱导愈伤组织,结果表明,叶柄是一种较好的外植体,在培养基MS+0.5 mg/L TDZ+0.2 mg/L NAA上,愈伤诱导率高达98.33%;将叶柄愈伤组织转接到培养基MS+0.2 mg/L IAA+1.0 mg/L 6-BA上进行分化培养,分化率高达86.67%,平均每块愈伤组织可以产生4.31个芽;组培苗在培养基1/2 MS+0.5 mg/L IAA上,最大生根率可达86.67%,平均每株幼苗可以产生5.24条根,平均根长达到4.31 cm,移栽成活率达100%。本研究为香果树种苗繁育及其遗传转化技术提供指导。Emmenopertys henryi Oliv.is a unique tree species in China,which is rare and endangered.To establish a regeneration system of high frequency for Emmenopertys henryi,the leaves and petioles of E.henryi were used as explants,and the new phytohormone TDZ and NAA were applied in the medium to induce callus.Results showed that petioles were better as explants.On medium MS+0.5 mg/L TDZ+0.2 mg/L NAA,the callus induction frequency reached 98.33%.Transferring the calli of petioles to medium MS+0.2 mg/L IAA+1.0 mg/L6-BA for differentiation culture,the shoot regeneration frequency was up to 86.67% with 4.31 shoots per callus.On medium 1/2 MS+0.5 mg/L IAA,the maximum root growing rate was 86.67%.Averagely,every shoot could produce 5.24 roots,and the average root length was 4.31 cm.The plantlets could be successfully planted in soil with a high survival rate of 100%.This study could provide guidance for the seedling breeding and genetic transformation technology of Emmenopterys henryi Oliv..
