摘  要:目的:研究五子衍宗方对自然衰老大鼠睾丸损伤的保护作用及其机制。方法:以SPF级雄性SD大鼠为研究对象,将18月龄大鼠随机分为自然衰老组及五子衍宗方低、中、高剂量组,每组10只。另以10只2月龄大鼠作为青年对照组。五子衍宗方低、中、高剂量组分别灌胃给药0.4、0.8、1.6 g/kg,青年对照组、自然衰老组灌胃生理盐水,每周停药2 d,连续给药4个月。末次给药后称量质量并处死大鼠,称取睾丸质量,计算睾丸指数。HE染色观察睾丸组织形态学变化,电镜观察睾丸支持细胞超微结构,生化法检测睾丸组织中SOD、MDA、GSH的水平,Western blot检测睾丸中增殖分化相关蛋白GDNF、SCF、BMP4表达。结果:与自然衰老组比较,五子衍宗方各剂量能不同程度升高自然衰老大鼠的睾丸质量和睾丸指数。HE结果显示,五子衍宗方能显著改善自然衰老大鼠睾丸组织形态结构;与青年对照组比较,自然衰老组大鼠睾丸组织SOD、GSH活性显著降低,MDA含量性显著升高,五子衍宗方干预后SOD、GSH活性不同程度升高,MDA含量不同程度降低。电镜结果显示,五子衍宗方能显著改善自然衰老大鼠睾丸Sertoli细胞的超微结构。与自然衰老组比较,五子衍宗方能显著上调自然衰老大鼠睾丸GDNF、SCF、BMP4蛋白表达。结论:五子衍宗方对自然衰老大鼠睾丸损伤有显著的保护作用,其作用机制可能与提高机体抗氧化能力,进而改善Sertoli细胞结构和功能有关。Objective:To study the protective effects of Wuzi Yanzong prescription on testis injury in natural aging rats,and to explore the possible mechanism. Methods:The 18-month old male SD rats were randomly divided into natural aging group,Wuzi Yanzong prescription low-, middle- and high- dose group, with ten rats in each group. Another ten two-month old rats used as young control group. Wuzi Yanzong prescription low-, middle- and high- dose groups were given gastric gavage of Wuzi Yanzong prescription 0. 4,0. 8, 1.6 g/kg, respectively,the young control group and the natural aging group were given normal saline and withdrawal 2 d a week,lasting for four months. The rats were sacrificed after the final treatment of Wuzi Yanzong prescription and then weighting body mass, and the testis were obtained to calculate the testieular weight and the testicular index. HE staining was used to observe the testicular tissue morphology ,the microstructure of sertoli cells was observed by Electron microscopy. The levels of SOD, MDA anti GSH in the testis were detected by biochemical methods,and the relative expression levels of GDNF,SCF, BMP4 in testieular tissue were detected by Western blot. Results : Compared with the natural aging group,the testicular weight and testicular index of natural aging rats increased significantly in Wuzi Yanzong prescription different doses groups. HE staining results showed that Wuzi Yanzong preseription could significantly improve the testicular tissue structure of natural aging rats. Compared with the young control group,the activity of SOD and GSH of rats testis in the natural aging group decreased significantly,the content of MDA increased significantly ;conversely ,the activity of SOD and GSH increased in varying degrees,and MDA content decreased in varying degrees alter Wuzi Yanzong prescription intervention.
