摘  要:目的探讨人参皂苷Rg1对糖尿病大鼠足创面愈合的影响及相关机制。方法 96只SD大鼠随机分为3组:糖尿病组(DM组)、人参皂苷Rg1(G-Rg1组)和阳性对照组。DM组和G-Rg1组大鼠采用腹腔注射链脲佐菌素的方式建立糖尿病模型,造模成功后,3组大鼠均进行足部创面造模。阳性对照组不予干预,正常愈合;G-Rg1组,按40 mg·kg^(-1)·d^(-1)的剂量予以G-Rg1生理盐水溶液灌胃;DM组,予生理盐水灌胃。分别于干预后3、7、14 d取创面组织,进行评价。结果糖尿病大鼠造模成功。HE和Masson染色表明,G-Rg1组和阳性对照组的上皮连接完好,新生胶原纤维和管腔组织均优于DM组。各组大鼠Col-1、Col-3及VEGF基因的表达趋势一致,均在术后前期达到高峰,术后14 d明显回落,组间比较显示,G-Rg1组和阳性对照组表达显著高于DM组,差异有统计学意义(P<0.05),但两组间差异无统计学意义。各组大鼠Col-1、Col-3及VEGF蛋白的表达均在术后前期达到高峰,术后14 d回落,Col-1及VEGF组间比较显示,G-Rg1组和阳性对照组表达显著高于DM组,差异有统计学意义(P<0.05),但两组间差异无统计学意义。Col-3的结果趋势基本一致,但术后7 d的阳性对照组显著优于G-Rg1组与DM组,差异有统计学意义(P<0.05),而G-Rg1组与DM组差异无统计学意义(P>0.05)。结论 G-Rg1可以通过上调VEGF和Col-1、Col-3的表达来促进新生血管和胶原蛋白的生成,从而改善糖尿病大鼠创面的修复。Objective To evaluate the effect and mechanism of ginsenoside-Rg1 on foot wound healing in Sprague-Dawley rat models with diabetic uleer. Methods 96 SD rats were divided into 3 groups: diabetes mellitus group (DM group), ginsenoside-Rgl group (G-Rgl group) and positive control group. After the diabetic rat models were established by a single intraperitoneal injection of streptozutoein (STZ) in DM group and G-Rgl group, 3mm ×7mm full-thickness skin defects were created on the feet of all rats. Normal wound healings were acquired in positive eontrnl gTuup with no intervention. G-Rgl saline solution was administrated by gavage at a dosage of 40mg·kg-1·d-1 in G-Rg1 group. Correspondingly, the rats were given normal saline of" the same quantity in DM group. Wound tissue samples were obtained and evaluated on 3 days, 7 slays and 14 slays after the interventions respectively. Results The diabetic rat models were successful. Histologically, HE anti Masson staining resuhs indicated that well-connected epithelium was observed in G-Rg1 and positive control groups, besides, new collagen fibril ansi tubular construction in G-Rgl and positive control groups were more than those in the DM group. PCR results revealed that the expression levels of Col- 1 , Col-3 and VEGF mRNA in the G-Rg1 and positive control groups were considerably up-regulated compared with those in the DM group at different time points ( all P〈0.05 ). The same trend was also observed in the protein expression of these cytokines detected by Western blot. Specifically, the expression level of Col-3 protein in the pnsitive control group was significantly superior to that in the G-Rgl and DM groups (P〈0.05), however, there is no signifieant difference between the two groups (P〉0.05).
