摘 要:目的对比研究金匮肾气丸加味膏方和汤方干预肾阳虚模型的疗效。方法将40只SD雄性大鼠分为正常组、模型组、金匮肾气丸加味膏方组(简称肾气丸膏方组)、金匮肾气丸加味汤方组(简称肾气丸汤方组),除正常组以外各组通过采用按200 mg/kg剂量灌服腺嘌呤4周的方法复制肾阳虚证大鼠模型,肾气丸膏方组和肾气丸汤方组分别给予18.36 g/(kg·d)的金匮肾气丸加味膏方和汤方,持续干预6周。第6周末,腹主动脉抽取各组大鼠动脉血,取血清,检测各组大鼠下丘脑-垂体-性腺轴激素如睾酮(T)、雌二醇(E2)、黄体生成素(LH)、人绒毛膜促性腺激素(HCG),下丘脑-垂体-肾上腺轴激素如促肾上腺皮质激素(ACTH)、皮质醇(COR)、17-羟皮质类固醇(17-OH-CS),下丘脑-垂体-甲状腺轴激素如三碘甲状腺原氨酸(T3)、四碘甲状腺原氨酸(T4)的含量。结果与正常组相比,模型组LH含量显著下降(P<0.05),肾气丸膏方组与肾气丸汤方组LH皆显著升高(P<0.05)。与正常组相比,模型组E2含量显著下降(P<0.05),肾气丸汤方组E2较模型组显著升高(P<0.05),且肾气丸汤方组E2含量显著高于膏方组(P<0.05)。与正常组相比,模型组T有下降的趋势,但无显著性差异,肾气丸膏方组和肾气丸汤方组T较模型组显著升高(P<0.05)。与正常组相比,模型组17-OH-CS含量显著升高(P<0.05),肾气丸汤方组较模型组含量显著下降(P<0.05),与肾气丸膏方组相比,肾气丸汤方组17-OH-CS含量降低显著(P<0.05)。与正常组相比,模型组T4含量显著下降(P<0.05),与模型组相比,肾气丸膏方组T4含量显著升高(P<0.05),与肾气丸汤方组相比,肾气丸膏方组T4含量显著升高(P<0.05)。结论金匮肾气丸加味膏方和汤方两种剂型均可以通过提高促LH和T含量起到温补肾阳的功效,但是两种剂型又有差异,汤剂可以通过提高E2含量和降低17-OH-CS起到温补肾阳的功效,而膏剂则对此两指标无显著影响;膏�Objective To compare and study the curative effects of modified paste and decoction of Jinhui Shenqi (Kidney-qi Pill in Synopsis of the Golden Chamber) Pill in intervention of rat model of kidney yang deficiency. Methods Male SD rats (n = 40 ) were divided into normal group, model group, group of Jinhui Shenqi Pill paste (paste group ) and group of Jinhui Shenqi Pill decoction (decoction group). Expect of normal group, other groups were orally given adenine (200 mg/kg) for 4 weeks for copying rat model of kidney yang deficiency pattern. The paste group and decoction group were respectively given modified paste or decoction of Jinhui Shenqi Pill for 6 weeks. The model group was orally given distilled water and ginseng group was given conventional breeding. On the end of the 6th week, the serum samples were collected from all groups for detecting the hormones of hypothalamus- pituitary-gonadal axis including testosterone (T), estradiol (E2), luteinizing hormone (LH) and human chorionic gonadotropin (HCG) , hormones of hypothalamus-pituitary-adrenal axis including adrenocortico- trophic hormone (ACTH), cortisol (COR), 17-hydroxycorticosteroid (17-OH-CS), and hormones of hypothalamus-pituitary-thyroid axis including tri-iodothyronine ( T3 ) and tetra-iodothyronine ( T4 ). Results The content of LH decreased significantly in model group compared with normal group (P 〈 0.05) , and increased significantly in paste group and decoction group (P 〈 0.05 ) but there was no significant difference between paste group and decoction group. The content of E2 decreased significantly in model group compared with normal group ( P 〈 0.05 ) , increased significantly in decoction group compared with model group (P 〈 0.05). There was no significant difference in content of E2 between paste group and model group, but it was significantly higher in decoction group than that in paste group (P 〈 0.05).
