银屑Ⅰ号抑制TNF-α诱导Hacat细胞炎症反应的机制研究

摘  要:目的:观察银屑I号对肿瘤坏死因子α(TNF-α)诱导的人永生化角质形成细胞(Hacat)炎症反应的影响并探讨其机制。方法:采用低、中、高浓度的银屑I号含药血清干预Hacat细胞,ELISA法检测细胞上清液25HVD3、IL-1、IL-17、IL-22浓度,Western-blot法检测维生素D受体(vitamine D receptor,VDR)蛋白表达量,RT-PCR检测VDRmRNA相对表达量。结果:与空白组比较,模型组VDR蛋白、VDRmRNA、25HVD3表达量明显降低,IL-1、IL-17、IL-22明显升高(P〈0.01);与模型组比较,银屑I号各组VDR蛋白、VDRmRNA、25HVD3表达量明显升高,IL-1、IL-17、IL-22明显降低(P〈0.05),且呈剂量依赖性。讨论:银屑I号可能通过促进VDR蛋白、VDRmRNA及25HVD3因子信号表达发挥对银屑病的治疗作用。Objective:To investigate the impact of Yinxie I Hao Decoction on the inflammatory response of Hacat cell model induced by tumor necrosis factor α and discuss its mechanism. Methods:Hacat cells were cultured by low,medium and high concentrations of Yinxie I Hao Decoction drug-containing serum,respectively. Enzyme-linked immunosorption test was inducted to detect the concentrations of 25 HVD3,IL-1,IL-17,IL-22 of supernatant fluid. The Western-blot was inducted to detect the expression of vitamin D receptor and the reverse transcription-polymerase chain reaction was inducted to detect the relative expression of VDRmRNA. Results:Compared with the blank group,the expressions of VDR protein,VDRmRNA and 25 HVD3 were decreased significantly and the concentrations of IL-1,IL-17 and IL-22 were increased obviously in model group( P〈0.01).Compared with the model group,the expressions of VDR protein,VDRmRNA and 25 HVD3 were increased significantly,while the concentrations of IL-1,IL-17 and IL-22 were decreased obviously in each dose groups of Yinxie I Hao( P〈0.05),and the influence was associated with concentration. Conclusion:Yinxie I Hao Decoction could improve the expressions of VDR protein,VDRmRNA and 25 HVD3 to antagonize inflammatory reaction.
