摘 要:目的:研究益气解毒复方对实验性自身免疫性重症肌无力(EAMG)大鼠B细胞趋化因子(CXCL13)、白细胞介素-21(IL-21)在血清及胸腺表达水平的影响。方法:100只Lewis大鼠中随机选取10只为正常组,其余采用对Lewis大鼠皮下注射鼠源性AchR-a亚基97-116肽段方法建立重症肌无力的实验模型,经评估造模成功的49只大鼠随机分为益气解毒复方高、中、低剂量组、醋酸泼尼松组、模型组。采用酶联免疫吸附试验(ELISA)法及免疫组化检测CXCL13、IL-21在EAMG大鼠血清及胸腺中的水平。结果:益气解毒复方高、中、低剂量组、醋酸泼尼松组血清及胸腺中CXCL13、IL-21水平明显降低,相比模型组有明显差异性(P〈0.01),但益气解毒复方高、中、低剂量组、醋酸泼尼松组之间比较无明显差异性(P〉0.05)。结论:益气解毒复方能明显改善EAMG大鼠肌无力症状,可降低EAMG大鼠血清及胸腺中CXCL13、IL-21水平,这可能是该方治疗MG的作用机制之一。Objective:To study the effect of Compound Yiqi Jiedu Decoction on CXCL13 and IL-21 levels in serum and thymus of rats with experimental autoimmune myasthenia gravis( EAMG). Methods:We selected 10 rats from 100 Lewis rats randomly as the normal group. The EAMG animal models were established in the rest 90 rats immunized by the rats' sequence 97-116 of the AChR a subunit( R97-116),49 of which were successful. According to the clinical scored,they were randomly divided into5 groups:Compound Yiqi Jiedu Decoction high,medium and low dose groups,prednisone group and model group. Enzyme linked immunosorbent assay( ELISA) and immunohistochemistry were used to detect the levels of CXCL13 and IL-21 in serum and thymus of EAMG rats. Results:The levels of CXCL13 and IL-21 in serum and thymus of Compound Yiqi Jiedu Decoction high,medium and low dose groups and prednisone group decreased significantly. There were significant differences compared with those of the model group( P〈0.01),but the Compound Yiqi Jiedu Decoction high,medium and low dose groups and prednisone group had no significant difference( P〉0.05). Conclusion:This prescription can obviously improve the symptoms of myasthenia gravis in EAMG rats,and can reduce the levels of CXCL13 and IL-21 in serum and thymus of EAMG rats,which may be one of the mechanisms of the treatment of MG.
