摘  要:目的观察白细胞介素13(interleukin-13,IL-13)刺激鼻息肉黏膜上皮细胞后嗜酸性趋化因子3,即CCL26蛋白含量及其mRNA表达变化,探讨嗜酸性粒细胞浸润鼻黏膜的机制。方法以IL-13细胞因子刺激原代培养的人嗜酸性鼻息肉黏膜上皮细胞。用实时定量PCR法检测CCL26 mRNA的表达,ELISA检测CCL26蛋白表达。结果与对照组相比,IL-13刺激组鼻息肉黏膜上皮细胞CCL26蛋白分泌增加,差异具有统计学意义(P<0.05)。其增高呈浓度依赖性。结论 IL-13可诱导鼻息肉黏膜上皮细胞分泌CCL26蛋白。OBJECTIVE To invest igate the changes of CCL26 protein content and its mRNA expression after stimulation of nasal polyps derived epithelial cells by Th2 cytokines, and to explore the mechanism of eosinophil inf iltration on nasal mucosa. METHODS Primar y cultured human eosinophilic nasal polyps derived nasal epithelial cells were stimulated with IL-13 cytokines. The expression of CCL26 mRNA was detected by real timePCR and the expression of CCL26 protein was detected by enzyme-linked immunosorbent assay(ELISA). RESULTS Compared with the control group, IL-13 stimulated the secretion of CCL26 protein from nasal epithelial cells. The difference was statistically significant(P〈 0.05, n=3). The increase is concentration-dependent. CONCLUSION Cytokine IL-13 can induce the secretion of CCL26 protein in nasal mucosal epithelial cells from nasal polyps.
