摘  要:为了将抗草甘膦基因EPSPS-G6导入棉花基因组中,以棉花品种中棉所49为受体材料,用草甘膦除草剂作为筛选剂,通过花粉管通道法最终获得了26个遗传转化事件,EPSPS-G6基因的PCR鉴定结果也同时表明目的基因已整合入棉花基因组。此外,15 mmol/L浓度草甘膦喷施后非转基因棉花品种TM-1和中棉所49全部死亡,表现为不抗草甘膦;而通过花粉管通道法获得的26个转化事件,对20 mmol/L浓度草甘膦均表现高抗,对40 mmol/L浓度草甘膦11个转化事件仍表现为高抗。In order to introduce the glyphosate resistant EPSPS-G6 gene into cotton genome, this study used the cultivar CCRI-49 as receptor parent and the glyphosate herbicide as screening agent. 26 genetic transformation events were obtained through the method of pollen tube pathway(PTP). At the same time, the results of PCR identification of EPSPS-G6 gene also indicated that the target gene was integrated into cotton genome.Furthermore, the non-transgenic cotton TM-1 and CCRI-49 both died after the treatment of 15 mmol/L glyphosate,which showed they were not resistant to glyphosate. However, all of 26 transformation events expressed a high resistance to 20 mmol/L glyphosate; 11 of them still expressed an excellent resistance to 40 mmol/L glyphosate.
