马缨杜鹃Actin基因片段的克隆及序列分析

摘　　要：为研究马缨杜鹃相关功能基因的表达模式提供内参基因,本研究根据Gen Bank中公布的锦绣杜鹃肌动蛋白基因(Actin)的保守序列设计引物,以马缨杜鹃c DNA为模板,利用RT-PCR方法克隆得到马缨杜鹃Actin基因部分c DNA片段,将其命名为Rd Actin。结果显示:该基因片段长度为468 bp,编码156个氨基酸;保守域分析表明该基因具有Actin的功能保守域,同时Rd Actin与其它植物Actin的氨基酸序列同源性达97%以上。In order to provide internal reference gene for the study of functional genes expression patterns of Rhododendron delavayi, we designed the primer on the basis of Aticn from Rhododendron x pulchrum in the Genebank. Using the cDNA from Rhododendron delavayi as template, we obtained Actin gene fragment by RT-PCR method, and named it as RdActin. The results showed that this Actin gene fragment was 468 bp, encoding a protein of 156 amino acids. Meanwhile, RdActin had the functional domains of Actin, and shared over 97% amino acid sequence homology with other plants actins.
